Molecular detection and phylogenetic analysis of partial sequence of Strawberry vein banding virus coat protein gene

Resumo: The cultivated strawberry, Fragaria x ananassa Duch. (Rosaceae) is a hybrid from the crossing of American species Fragaria chiloensis and Fragaria virginiana, belonging to the Rosaceae family. Strawberry plants have vegetative reproduction and, therefore, it is common the propagation of viruses and other diseases. One of the four most important viruses in strawberry is the Strawberry vein banding virus (SVBV), which is transmitted by aphids in a semi-persistent manner. Molecular techniques are recommended to improve genome knowledge and classification of the SVBV isolates. The coat protein (CP) is a primary determinant of aphid transmissibility and specificity, critically important to infection establishment. In the present work, we sequenced the SVBV coat protein partial gene of a German strain, inoculated in strawberry plants maintained in green house in Brazil for ten years. It was performed phylogenetic analysis comparing to SVBV coat protein sequences from the GenBank database. Aiming to elucidate evolutionary relationships in this species, phylogenetic analysis showed that our SVBV sequence is closely related to USA and Egypt virus strains. These results will contribute to a better elucidation of evolutionary mechanisms and the SVBV-strawberry pathosystem in question.

(Rosaceae) é um híbrido originado pelo cruzamento das espécies americanas Fragaria chiloensis e Fragaria virginiana e pertence à família Rosaceae. O morangueiro possui reprodução vegetativa e, por isso, é comum o acúmulo de viroses e outras doenças de difícil controle. Uma das quatro viroses mais importantes na cultura do morango é causada pelo vírus da faixa das nervuras (Strawberry vein banding virus – SVBV), que é transmitido no campo por afídeos, de maneira semipersistente. Para melhorar o conhecimento genômico, são recomendadas técnicas moleculares e a classificação de isolados de SVBV. Um dos determinantes primários da transmissibilidade e especificidade por afídeos é a proteína do capsídeo, que possui importância crítica para o estabelecimento da infecção. Neste trabalho, foi sequenciada parcialmente a proteína do capsídeo do gene do SVBV de um isolado alemão, inoculado em morangueiros e mantido em casa de vegetação no Brasil, por mais de dez anos. Foi realizada a análise filogenética comparando as sequências contidas no GenBank com o objetivo de elucidar as relações evolutivas nesta espécie. As análises filogenéticas mostraram que a sequência do isolado está mais próxima dos isolados dos EUA e Egito.
