Bioprospection of herbicide-resistant microorganisms

Resumo:   Herbicides are efficient tools for controlling weeds in crops; however there are many environmentalconcerns associated with their use. Scientists are developing strategies to better understand the relationships between herbicide use and damage to the environment. The present work aimed to isolate microorganisms with genes for resistance to, or degradation of the herbicides utilized in rice fields. Two experiments were carriedout: 1) to verify the presence of microorganisms in a tank where herbicides, insecticides and other agricultural products are discharged; 2) to verify the presence of microorganisms in flasks of the herbicide Nominee®. In thislast experiment, the capacity of the microorganisms to use the herbicide as their only carbon source was also tested. The culture media nutrient-agar (NA), Sabouraud and minimal medium (MM) were used. In experiment 1, the presence of microorganisms was verified in the discard tank. In experiment 2, microrganisms were isolated from a new and a used flask of the herbicide. The capacity of the microorganisms to use the herbicideas a source of carbon was proven. The strains isolated in this work were purified and addressed for experiments to test their resistance to other herbicides, as well as the capacity to degrade them.

O uso de herbicidas facilita o controle das plantas daninhas, mas preocupa quando o assunto é impacto ambiental. Por isso, muitos cientistas vêm somando forças para entender mais sobre os herbicidas e as suas relações com o ambiente. Foi realizado um estudo com o objetivo de isolar microrganismos com genes de resistência e/ou degradação de herbicidas utilizados na cultura do arroz. Foram realizados dois experimentos: 1) verificação da presença de microrganismos em um tanque de descarte de agrotóxicos; 2)isolamento de microrganismos de dois frascos do herbicida Bispyribac-sodium (Nominee®), um novo e outro em uso, e a capacidade desses microrganismos usarem o agrotóxico como única fonte de carbono. Foram utilizados os meios de cultura nutriente-ágar (NA), Sabouraud e meio mínimo (MM). No experimento 1 constatou-se a presença de microrganismos no tanque de descarte. No experimento 2 foram isolados microrganismos do interior dos dois frascos de herbicida testados. Ainda nesse experimento, observou-se a capacidade de utilização do herbicida pelos microrganismos. As cepas isoladas neste trabalho foram purificadas e direcionadas para testes de resistência e/ou degradação de outros agrotóxicos.
