Use of the formalin test to evaluate the antinociceptive potential of lemongrass essential oil (Cymbopogon citratus) using the fish Danio rerio

Resumo: Pain is characterized as an unpleasant sensory and emotional experience associated with tissue damage or injury, being detected through specialized receptors called nociceptors. Antinociception is the process where the transmission of nociceptive information is inhibited, being mediated by endogenous systems and achieved by exogenous substances, such as drugs. The research for new pharmacological compounds for pain treatment is a wide field of study, therefore, the present work aimed evaluate a new biological model using the fish D. rerio to analyze the antinociceptive potential of substances extracted from plants. Twenty-four fish were filmed for 5 min (baseline), treated with vehicle (Control, n=8), C. citratus essential oil (200 mg/kg, n=8) or dipyrone (100 mg/kg, n=8), after 30 minutes the animals were submitted to the formalin nociceptive test and filmed for 5 min (post-stimulus) for behavioral evaluation. Decreased locomotor activity was observed in response to formalin, and this effect was blocked by treatment with dipyrone. The essential oil (EO) did not present significant effects on nociception, requiring further studies to determine its analgesic potential.

A antinocicepção é o processo em que há a inibição da transmissão da informação nociceptiva, sendo mediada por sistemas endógenos e alcançada por substâncias exógenas, como os fármacos. A necessidade de se buscar novos compostos farmacológicos que auxiliam no tratamento da dor compõe um amplo campo de estudo, sendo assim, o presente trabalho objetivou testar um modelo biológico utilizando o peixe D. rerio para avaliação do potencial antinociceptivo de substâncias extraídas de plantas. Utilizou-se 24 peixes que foram filmados por 5 min (linha de base), tratados com veículo (Controle, n=8), óleo essencial de C. citratus (200 mg/kg, n=8) ou dipirona (100 mg/kg, n=8), após 30 minutos os animais foram submetidos ao teste nociceptivo da formalina e novamente filmados por 5 min (pós-estímulo), para avaliação comportamental. Foi observada diminuição da atividade locomotora em resposta à formalina, sendo este efeito bloqueado pelo tratamento com dipirona.
