Molecular bioprospecting of plant extracts: Experience report of the BIOPROS/UFV group in the search for antitumor compounds

Resumo: This manuscript discusses the experience of the research group BIOPROS (Molecular Bioprospecting in the Sustainable Use of Biodiversity) from the Federal University of Viçosa in the field of bioprospecting. We describe our experience on the search for antitumor compounds from the collection of extracts of native tree species from the Atlantic Forest biome. Presenting an interdisciplinary approach, integrating knowledge of forestry engineering, bioinformatics and natural products chemistry, the bioprospecting research of the BIOPROS group has innovated in the generation of pharmacochemical knowledge of native species of the Atlantic Forest. For the composition of the extract library a total of 220 plant species distributed in 57 botanical families and 140 genera were identified. 196 extracts from 49 plant species were produced; all of them evaluated for cytotoxic activity. By showing the process of obtaining a promising antitumor activity withanolides compounds from Athenaea velutina, a species hitherto little known to science, this manuscript shows our sequence of methodological steps used to unravel bioactive natural products from fragments of Atlantic Forest. The research follows the premises of the Convention on Biological Diversity, regarding the creation of strategies for the sustainable use of biodiversity.

El trabajo presenta un relato de experiencia en la búsqueda de compuestos antitumorales a partir de extractos de especies arbóreas nativas de la Mata Atlántica. Con un enfoque interdisciplinario, integrando conocimientos de ingeniería forestal, bioinformática y química de productos naturales, la investigación en bioprospección del grupo BIOPROS ha ido innovando en la generación de conocimiento farmacoquímico de especies de la Mata Atlántica. Para la recolección de extractos, se identificaron un total de 220 especies de plantas distribuidas en 57 familias botánicas y 140 géneros. En la búsqueda de compuestos antitumorales, se produjeron 196 extractos de 49 especies de plantas; siendo todos los extractos evaluados para actividad citotóxica. Al mostrar el caso de obtención de compuestos de withanólidos, con prometedora actividad antitumoral, a partir de una especie hasta ahora poco conocida por la ciencia, Athenaea velutina, el manuscrito muestra la secuencia de pasos metodológicos utilizados para llegar a productos naturales bioactivos . La investigación sigue las premisas del Convenio sobre la Diversidad Biológica, en cuanto a la creación de estrategias para el uso sostenible de la biodiversidad.O presente manuscrito diz respeito à experiência do grupo de pesquisa BIOPROS (Bioprospecção Molecular no Uso Sustentável da Biodiversidade), da Universidade Federal de Viçosa, no campo da bioprospecção. O trabalho traz um relato de experiência na busca de compostos antitumorais oriundo  de extratos de espécies arbóreas nativas da Mata Atlântica. Apresentando uma abordagem interdisciplinar, integrando conhecimentos da engenharia florestal, bioinformática e de química de produtos naturais, a pesquisa em bioprospecção do grupo BIOPROS tem inovado na geração de conhecimento farmacoquímico de espécies da Mata Atlântica. Para a coleção de extratos, um total de 220 espécies de plantas distribuídas em 57 famílias botânicas e 140 gêneros foram identificadas. Na busca de compostos antitumorais, foram produzidos 196 extratos oriundos de 49 espécies vegetais; sendo todos os extratos avaliados quanto atividade citotóxica. Ao mostrar o caso de obtenção de compostos withanolideos, com promissora atividade antitumoral, a partir de uma espécie até então muito pouco conhecida da ciência, Athenaea velutina, o manuscrito mostra a sequência de etapas metodológicas empregadas para se chegar a produtos naturais bioativos oriundos de fragmentos de Mata Atlântica.
