Resumo: | Integrins are important agents in communication between the cytoskeleton and plasma proteins or extracellular matrix intermediating cell-to-cell adhesion with other membrane proteins. Besides the adhesion function, integrin are recognized signaling molecules capable of performing transduction messages by classical pathways signaling thanks to their molecular structure. Thus, the integrins are consider as key modulators of cellular proliferation, migration, longevity, motility, and further, keep specific functions of cell differentiation. This review presents pertinent information about integrin complexity in cell signaling pathways, adhesion, migration and a key role in the hematopoietic system, especially the immune system. As a complex system, any failure provides greater opportunities for pathologies development. Thus, anomalies in integrinmediated signaling might contribute to different disease states, metastasis, angiogenesis and inflammatory diseases. Finally, some antagonists of integrins that are involved in clinical trials and new therapies are exemplified. |

Além da função de adesão, as integrinas são reconhecidas moléculas de sinalização capazes de realizar a transdução de mensagens por vias de sinalização clássica, graças a sua estrutura molecular. Desse modo, as integrinas podem ser consideradas moduladores chave da proliferação, migração, longevidade, migração e ainda, a manutenção das funções específicas de diferenciação celular. Esta revisão aborda informações pertinentes de como as integrinas funcionam de modo complexo dentro de vias de sinalização, de adesão, do citoesqueleto e desempenham papel fundamental no sistema hematopoiético, especialmente no sistema imune. Como sistema complexo, qualquer falha oferece maiores oportunidades para o desenvolvimento de patologias. Desse modo, aberrações na sinalização mediada pelas integrinas podem contribuir para diferentes estados patológicos, como metástase, angiogênese e doenças inflamatórias.
