Effect of therapeutic ultrasound in pulse mode on the release, retention and permeation of ascorbic acid in a vertical diffusion system

Resumo: Transdermal drug delivery offers many advantages over other routes of administration, such as potential for superior efficacy and minimal side effects. However, ways are sought new to increase the permeation of these active ingredients in the skin. A commonly used resource for this purpose is the phonophoresis; however, the real effectiveness of phonophoresis in numerous topical substances, such as ascorbic acid, is still unknown. In this sense, this study compares the effect of 5% ascorbic acid release, retention and permeation associated with ultrasound from studies in vitro. The release analysis was performed at 5, 10, 15 and 20 minutes, and the retention and permeation analysis was used in pig skin at 20 minutes, 1 and 2 hours. It was observed that in the release assays the ultrasound had the ability to increase the release of the active in the times of 5 and 10 minutes. In relation to the retention tests, the ultrasound was effective when compared to the control group in a time of 1 hour for stratum corneum and epidermis, given the remainder of the analyzed times it was not possible to analyze the difference among the analyzed groups.

Entretanto, buscam-se novas formas de ampliar a permeação desses fármacos na pele. Um recurso utilizado para essa finalidade é a fonoforese, que através do Ultrassom (US), gera uma onda acústica, também chamada de cavitação, sendo capaz de aumentar a permeação de princípios ativos sobre a pele. O estudo compara o efeito da liberação, retenção e permeação do ácido ascórbico (AA) 5% associada ao US. Em análise de liberação utilizou-se ácido ascórbico com US nos tempos de 5, 10, 15 e 20 minutos. Em análise da retenção e permeação utilizou-se pele de suíno nos tempos de 20 minutos, 1 e 2 horas. Observou-se diferença na liberação da substância, porém, quando comparado com os tempos utilizando o US houve diferença nos tempos de 5 e 10 min.
