Cell cycle: regulatory mechanisms and biochemical markers

Resumo: The regulatory mechanisms of the cellular cycle and its relationship with the growth factors, oncogenes end anti-oncogenes are one of the major advanced fields in the cancerbiology studies. The Identification of proteins and cofactors regulating the transitions between G1/S and G1/M as well the transcription of the genetic material have allowed the detection of cyclíng cells even before the appearance of the mitotic signs. The development of techniques using monoclonal antibodies against those proteins have made possible studies on proliferative cells of different tomours, mainly based on fact that proteins such as alpha polymerase and cyclin/PCNA are expressed only by proliferating cells. Thus the detection and counting of these cells would allow the evaluation of either the tumor-growth rate or its therapeutic efficiency and consequentiy the prognostic of the disease. The present paper reviewed the current knowledgement of the cell-cycle control mechanisms and the available methodology for the proliferative-cell detection emphasizing the biochemical markers, its practicability and reliability.

Continuamente estão sendo descobertas novas proteínas envolvidas no complexo mecanismo que desencadeia as diferentes fases do ciclo celular, como também os seus mecanismos de ativação. A descoberta dessas proteínas e co-fatores que regulam as transições G1/S e G2/M e que atuam na replicação do material bem como o papel dos proto-oncogenes dentro do ciclo celular permitiu aumentar o conhecimento sobre as células ciclizantes como tarnbém, permitiu que estas pudessem ser detectadas mesmo sem a visualização de figuras mitóticas. O desenvolvimento de anticorpos monoclonais contra estas proteínas tornou viável o estudo, em larga escala, de taxas de proliferação celular em diferentes neoplaslas, uma vez que substâncias como Ki67, polimerase alfa e PCNA - Ciclina somente são expressas em células em proliferação. A detecção de células proliferativas permite, através de sua contagem, estimar as taxas do crescimento neoplásico com conseqüente repercussão no prognóstico da doença e na terapêutica do paciente.
