Resumo: The availability of complete and partial genomes of prokaryotes has increased exponentially in public biological databases due to advances in genome sequencing techniques in recent years. Two years ago, after the installation of an infrastructure for DNA sequencing, the Bioinformatics Laboratory of the National Laboratory for Scientific Computation entered this era of large-scale sequencing. The in-silico genome analysis using these sequences was produced with very different purposes, such as comparative analysis of genomes, studies of metabolic pathways, phylogenetic analysis, among others. The larger supply of phylogenetically close genomes may facilitate biological inferences, but requires the development of tools to facilitate and accelerate the annotation and comparison of genes. The System for the Comparison of Prokaryotic Genomes - SCGP groups all possible orthologous genes given a set of genomes, stores them in a database as ix well as allows queries of genomes selected (by some annotation data such as product, gene name, synonym and amino acid sequence). From the clusters generated the system can infer information about common genes between the genomes studied, gene order and functional classification. The system also allows easy updating and adding new genomes from a previously comparison. For validation of the system, 19 genomes from the order Mycoplasmatales were tested. It was possible to search information about similarity and sequence alignment. On the other hand, the analysis method from the clusters allowed access to the list of homologous sequences groups for identifying the possible evolutionary relationships, structure as well as function between the genomes studied.

Há dois anos, após a instalação de uma infra-estrutura de sequenciamento de DNA, o Laboratório de Bioinformática do Laboratório Nacional de Computação Científica ingressou nessa era de sequenciamento em grande escala. As análises genômicas in-silico utilizam-se dessas sequências produzidas com os mais diversos fins, como, por exemplo, a análise comparativa de genomas, estudos de vias metabólicas, análises filogenéticas dentre outras. Soma-se também ao fato de que a maior oferta de genomas de organismos filogeneticamente próximos pode facilitar inferências biológicas, mas demanda o desenvolvimento de ferramentas que facilitem e agilizem a anotação e a comparação dos genes. vii O SCGP - Sistema de Comparação de Genomas de Procariotos agrupa todos os possíveis genes ortólogos a partir de um conjunto de genomas, armazena- os em um banco de dados, permite consultas de genomas selecionados, utilizando dados da anotação como produto, nome de gene, sinônimo e sequência de aminoácidos. A partir dos clusters gerados é possível inferir informações sobre genes comuns entre os genomas estudados, ordem gênica e classificação funcional. O sistema ainda permite fácil atualização, adicionando novos genomas em uma comparação já existente. Para validação do sistema foram usados 19 genomas da ordem Mycoplasmatales.
