Resumo: The acute respiratory distress syndrome (ARDS) is a severe lung disease, multifactorial, characterized by diffuse alveolar damage, infiltration of inflammatory cells, disruption of barrier of the alveolar epithelium, alveolar edema and impaired pulmonary gas exchange with high index of mortality and absence of efficient treatment and also, sepsis is the most common cause. The mechanisms involved in acute lung disease induced by systemic inflammation are investigated experimentally by administration of LPS systemically. This model develops inflammatory lesion with pulmonary neutrophilic infiltration and increased release of cytokines, including IL-1β and TNF- . The photobiomodulation has shown good results for various diseases with inflammatory character; however, there are few studies related with lung diseases. Considering that ARDS is an important pathology, we studied the effects of LED treatment on the development of ARDS. For this purpose, groups of mice were treated with LPS for ARDS induction and treated or not with LED (trachea and lungs) 2 and 6 h after the LPS injection. In parallel, non-manipulated group as well as treated only with LED were used as control. After 24h of LPS administration, we investigated the number of cells recruited in bronchoalveolar lavage (BAL), blood and bone marrow, quantification of cytokines in the BAL supernatant as well as evaluation of myeloperoxidase enzyme (MPO), oxidative burst, phagocytosis and histology. Our results indicated the beneficial effects of LED treatment on the ARDS model induced by LPS, since the treatment with LED significantly reduced the number of neutrophils recruited to the lung, MPO activity, oxidative burst, and also reduces the release of IL- 1β, IL-6 and TNF-α concomitantly with the increased levels of IFN-γ and IL-10 gene expression. Therefore, this study may provide important information to treatment of ARDS.

Therefore, this study may provide important information to treatment of ARDS.A Síndrome do Desconforto Respiratório Agudo (SDRA) é uma patologia pulmonar grave, de causa multifatorial, caracterizada por lesão alveolar difusa, infiltrado de células inflamatórias, ruptura na barreira do epitélio alveolar, edema alveolar e comprometimento da troca gasosa pulmonar, com alto índice de mortalidade e sem cura até os dias atuais, sendo a sepse sua causa mais comum. Os mecanismos subjacentes à inflamação pulmonar aguda decorrente da inflamação sistêmica são investigados experimentalmente pela administração de lipopolissacarídeo (LPS) por via sistêmica. Esse modelo desenvolve lesão inflamatória com infiltração neutrofílica pulmonar e é potencializada por diversas citocinas, entre as quais se destacam IL-1β e TNF- fotobiomodulação vem apresentando bons resultados para diversas doenças com caráter inflamatório, porém, ainda não existem muitos estudos relacionando o uso do LED no tratamento de doenças pulmonares. Considerando que a SDRA é uma importante patologia, estudamos o efeito do LED sobre o seu desencadeamento. Para tanto, grupos de camundongos foram submetidos a SDRA através da injeção de LPS (ip) e tratados ou não com LED na região da traquéia e pulmão 2 e 6 h após a indução de SDRA com LPS. Paralelamente, grupos de camundongos não manipulados e grupo de camundongos apenas tratados com LED foram utilizados para a obtenção dos valores basais. Após 24 h da indução de SDRA foram analisados o número de células em diferentes compartimentos (lavado broncoalveolar (LBA), sangue e medula óssea), expressão e quantificação e de citocinas, avaliação da enzima mieloperoxidase (MPO), burst oxidativo, fagocitose e histologia. Nossos resultados apontam para os efeitos benéficos do tratamento com LED sobre modelo de SDRA induzida por LPS, uma vez que reduziu significativamente o número de neutrófilos recrutados para o pulmão, a atividade de MPO e burst oxidativo, também reduziu a liberação de IL-1β, IL-6 e TNF- α, concomitantemente ao aumento de IFN-γ e a expressão de IL-10.
