Nos últimos anos, a neuroinflamação foi proposta como uma das características chave na DA, com potencial impacto no aumento da acumulação de Aβ e emaranhados neurofibrilares. Um aspeto importante da patologia de DA, é o envolvimento de diferentes regiões do cérebro. O hipocampo é uma região cerebral responsável pela consolidação de memória a curto prazo para memória a longo prazo, sendo uma das áreas mais afetadas na DA. Diferentes análises lipidómicas demonstraram o papel preponderante dos lípidos no cérebro e também o envolvimento destes na DA. Os cérebros de pacientes com DA mostraram ter um elevado conteúdo de inclusões adiposas, sugerindo um desequilíbrio no metabolismo lipídico. Foi também demonstrado que a atividade da fosfolipase D, responsável por hidrolisar fosfatidilcolina em ácido fosfatídico, está alterada. A nossa equipa, demonstrou anteriormente que a proteína Aβ provoca um aumento na atividade da fosfolipase D em culturas de neurónios primários e que a ablação genética da isoenzima fosfolipase D2 atenua a disfunção sináptica e os défices cognitivos num modelo de murganho com DA. A via da fosfolipase D está envolvida no normal funcionamento do hipocampo e indivíduos com DA demonstraram ter a sua atividade alterada. Assim, a nossa hipótese é que a ablação genética da fosfolipase D2 poderá alterar fenótipos característicos da DA, através da modulação da neuroinflamação. O presente estudo pretende perceber de que forma a ablação da fosfolipase D2 poderá alterar a resposta inflamatória no hipocampo de murganhos J20, um modelo utilizado para estudar a DA. Através de análises histológicas e bioquímicas, foi possível verificar que a percentagem de área coberta por células de microglia está reduzida em todas as sub-regiões do hipocampo dorsal e ventral, verificando-se também a existência de alterações na morfologia destas células em direção a um estado amoebóide. Os nossos resultados apontam também, para possíveis alterações morfológicas nos astrócitos após a ablação da fosfolipase D2. Assim, o nosso estudo sugere que a ablação da fosfolipase D2 promove alterações na ativação das células da glia do hipocampo, tanto em murganhos não transgénicos como murganhos J20, indicando que os efeitos na resposta inflamatória cerebral provocados pela fosfolipase D2 podem ir além da acumulação de Aβ no cérebro.Alzheimer’s Disease (AD) is the most common form of dementia, and it is characterized by the brain accumulation of amyloid beta (Aβ) peptides, giving rise to neuritic plaques and intracellular neurofibrillary tangles (NFTs), which are constituted by hyperphosphorylated tau. Over the past years, neuroinflammation has been proposed as a third core feature of AD, with potential impact in exacerbating Aβ burden and NFTs. Another important aspect of AD pathology is the differential involvement of specific brain regions. The hippocampus, a brain region responsible for the consolidation of short-term memory into long-term memory, is one of the most affected regions in AD. Several lipidomic analysis showed that lipids are key regulators of brain function and have additionally been implicated in AD. Indeed, brains from AD patients display higher content of ‘adipose inclusions’ suggesting lipid dyshomeostasis. In accordance with these studies, phospholipase D (PLD) activity, that hydrolyses phosphatidylcholine (PC) to generate phosphatidic acid (PA), was shown to be disturbed. Importantly, our team previously showed that Aβ leads to an increase in PLD activity in primary neuronal cultures and that the genetic ablation of the isoenzyme phospholipase D2 (PLD2) ameliorates synaptic dysfunction and cognitive deficits in AD mice. The PLD pathway has been implicated as a key determinant of the normal function of the hippocampus and, AD individuals display altered PLD metabolism. We then hypothesized that the genetic ablation of PLD2 could impact AD-phenotypes through neuroinflammation modulation. Therefore, the main goal of our study was to assess how PLD2 ablation affect the inflammatory reaction in the hippocampus in J20-line AD mice. Through histological and biochemical analysis, we saw that the percentage of area covered by microglia is reduced in all hippocampal subfields both in dorsal and ventral hippocampus and that, microglia cells exhibit a morphology towards an amoeboid state, Moreover, although not statistically significant, our results indicate that astrocytes could also be displaying alterations in their morphological phenotypes upon PLD2 ablation.
