Lipidic Panel – Microfluidic disc biochemical functionalization

Resumo: | biosurfit is a Portuguese company focused on the development of in vitro diagnostic tests based on point-of-care technology (PoC). Carrying the challenge of decentralizing health systems,which are meant to be closer and personalized to the needs of patients, PoC tests (PoCT) are positioned as a strategic solution in diagnosis and monitoring. Consequently, this company developed spinit®, a fully automated diagnostic platform capable of performing several diagnostic tests on the same equipment, in minutes and with only a drop of blood. The spinit® technology comprises the spinit® equipment, which includes multiple detection methods (including spectrometry) and disposables that combine microfluidic centrifugation systems with the reagentes necessary for the biological processing of the sample. The company is currently developing a PoCT for the lipid panel - spinit® Lipids. Cardiovascular diseases generate approximately 18 million deaths per year globally mainly due to atherosclerosis. As cholesterol and triglycerides were appointed as markers for cardiovascular disease risk, a lipidic profile could be very elucidative to provide health professionals the necessary information and consequently improve patient health. spinit® Lipids disc is functionalized with enzymatic formulas, prepared using commercial quantitation kits and an excipient to protect the enzymes, which enables the quantification of TAGs, total cholesterol (TC) and high-density lipoprotein cholesterol (HDL-C) through colorimetric reactions. All other parameters shall be calculated, including and low-density lipoprotein cholesterol (LDL-C). The main goal of the present thesis is to assess different formulas and evaluate several performance requisites including their capacity to distinguish samples with different analyte concentration, posteriorly to production of the disposable test (dynamic range) and stability for long periods of time (shelf-life assessment).From the obtained results it was not possible to appoint the final formulas to be used in spinit® Lipids, yet a risk assessment was carried out for each one and future developments in the test were envisaged. |
