Universidade de LisboaPucciniales (Basidiomycota) is an order containing thousands of species, all of them biotrophic plant pathogens, establishing highly specific interactions with their hosts, frequently undergoing complex life cycles. In contrast with neighbouring orders, Pucciniales fungi present large genomes. Whereas diploid nuclei would be expectable only in basidia, in the present work the occurrence of haploid and diploid nuclei in a wide range of Pucciniales species across their life cycles is reported not only using FCM but also with FISH using rDNA probes, even if it was also detected in one fungus of the order Platygloeales the phenomenon is likely to be circumscribed to Pucciniales. Although all rust fungi are dikaryotic during the most part of their life cycles, the fungus Ctss presents nuclei with distinct genome sizes (the Twin Peaks phenomenon), and this trait allows the separation of the two nuclei populations by FACS. This constitutes an unprecedented tool for the study of the heterokaryon in Pucciniales. In the present work, a RNA-seq analyses was done on these nuclei populations (A and B) separately to evaluate the transcripts, allowing to conclude that 41% are common in both nuclei populations and ~33% are exclusive of A and 25% of B nuclei population. Jointly with the amplification of some DNA fragments of genes of interest by PCR it was proved that the populations of nuclei A and B effectively constitute the heterokaryon of Ctss. The commercial globalization, the environmental changes, and the appearance of new rust fungi in new locations enhance the need of detailed knowledge on rust-host interactions.
