A sua produção atual em sistemas biológicos não atinge um rendimento suficientemente alto para que seja economicamente viável. A levedura Saccharomyces cerevisiae é um modelo genético e bioquímico amplamente utilizado, assim como um organismo de produção industrial. A sua produção de FA está associada a um complexo proteico multifuncional, codificado pelos genes FAS1 e FAS2. Em outros organismos, como Escherichia coli e Arabidopsis thaliana, a mesma via é catalisada por enzimas monofuncionais expressas por genes únicos. Uma via monofuncional híbrida de E. coli e A. thaliana expressando doze genes individuais fora previamente desenvolvida, sendo o objetivo deste trabalho a investigação das consequências fisiológicas da substituição dos genes nativos FAS1 ou FAS2 pela mesma. As leveduras recombinantes foram sujeitas a um processo de adaptação em vários meios sem FA adicionados, assim como cada unidade genética fora explorada pela introdução de cópias adicionais de genes da via. Os perfis lipídicos foram determinados por fluorometria, gravimetria e cromatografia gasosa. As estirpes dependentes da via híbrida para a síntese de FA cresceram a um ritmo lento, com uma fase de latência extensa em comparação com a estirpe controlo. Em termos lipídicos, houve uma redução de 7 vezes no ácido Oleico em comparação com o sistema nativo. A adaptação das estirpes proporcionou um crescimento consideravelmente mais rápido e uma fase de latência reduzida. Cópias adicionais dos genes fabH de E.coli e mod1 de A. thaliana resultaram numa maior produção de C14 e a diminuição da saturação face aos determinados pela estirpe controlo.The urgency in substituting fossil fuels has triggered a growing interest in renewable biomolecules, such as medium-chain fatty acids (FA). Current fatty acid production in biological systems does not reach high enough titers for this to be economically viable. The yeast Saccharomyces cerevisiae is a widely used genetic and biochemical model and an industrial production organism. Yeast FA pathway is associated with a multifunctional protein complex, encoded by the FAS1 and FAS2 genes. In other organisms such as Escherichia coli and Arabidopsis thaliana, the same pathway is catalyzed by monofunctional enzymes expressed by single genes. A hybrid E. coli and A. thaliana monofunctional pathway expressing twelve individual genes had been previously established and this work aimed at investigating the physiological consequences of replacing the function of native FAS1 and FAS2 genes with this pathway. Recombinant yeast strains were subjected to adaptation on media without exogenous FA and gene dosage was explored by adding additional copies of individual pathway genes. Lipid profiles were determined by fluorometry, gravimetry and gas chromatography. Strains relying on the hybrid pathway for fatty acid synthesis grew at a low rate and with a prolonged lag phase compared to the control strain. In terms of lipids, a 7-fold reduction in Oleic acid compared to those with the native system. Strain adaptation resulted in strains that grew considerably faster and with a reduced lag phase.
