E-mail: bmsantos@ufv.br Three clones secreting of antibodies (Abs) IgG against infection bursal disease virus IBDV was development. The IBDV was strain S706 (the intermediate vaccine) was replicated in VERO cell and purified by sucrose gradient, for ELISA and mice inoculation. For the immunization of the mice BALB/c using as a saponin adjuvant, that allowed an inflammation reaction which enhanced the antibody response, detectable by ELISA. The fusion of splenic cells of the immunized mice and the mieloma SP2/0 resulted in 2 hybridoma families (2H11 and 5C7). After cloning by limiting dilution, 3 clones secretors of Abs from IgG class were obtained. The 3 obtained Abs were capable to reveal the proteins turn VPX and VP2 by “western blotting”, respectively of 47 kDa and 41 kDa. The definition of the isotypes recognized by obtained Abs must be object of characterization to allow the use of the antibodies in immunodiagnostic tests such as immunofluorescence, immunocitochemistry or capture ELISA, for epidemiologic of the disease researches or to differentiate vaccine’s virus of the field virus. |

Para a produção dos anticorpos (Ac) utilizou-se de um vírus inteiro proveniente de uma vacina comercial, estirpe intermediaria (Bur-706), que foi multiplicado em células VERO e purificado por ultracentrifugação em gradiente de sacarose, sendo utilizado para a imunização dos camundongos BALB/c. O adjuvante escolhido foi a saponina, que permitiu a obtenção de anticorpos policlonais capazes de revelar a enzima por ELISA. A fusão das células esplênicas dos camundongos imunizados e o mieloma SP2/0 resultou na obtenção de 2 famílias de hibridomas (2H11 e 5C7) secretores de anticorpos. Após clonagem por diluição limitante, foram obtidos 3 clones secretores de Ac da classe IgG. Os três Ac obtidos foram capazes de revelar as proteínas virais VPX e VP2 por ”western blotting”, reconhecendo a proteína de 47 kDa e 41 kDa e outra proteína não identificada com peso molecular de 66 kDa.
