Resumo: The autonomic nervous system (ANS) regulate and coordinate processes and function of organs to maintain homeostasis of the cardiovascular system in response of changes in the internal environment. Frequently, the understanding of the response patterns, interaction reflexes and other factors regulated by ANS, required in vivo studies. The use of anesthetics during the course of experimental protocols and data collection has been a way of solving the problems related of such studies. However, anesthetics depress the nervous system and compromise the conclusions from investigations that rely on functional ANS. Thus, in this work, we sought to validate a decerebrated preparation with individuals of the species Crotalus durissus. Decerebration consists in the ablation of superior brain areas in order to maintain preparations that are incapable of superior processing, perception and sensations of any type, including stress or pain. In these preparations, areas of the brainstem and part of the hypothalamus are maintained. Thus, the cardiovascular homeostasis is maintained. In the present study, we performed series of physiological tests for validated the presence of autonomic responses in the model. In addition, we performed a baroreflex study to understand how effective is the model in representing the intact animal. Decerebrated animals were able to keep hemodynamic variables constant during the tail lift protocol. In the head lift maneuver, the systemic mean arterial pressure (MAP sys) increased to ensure adequate blood supply to the head. In addition, during surface heat application, the heart rate (fH) increased, a cardiovascular response typical of Squamata group members during heating. Pharmacological stimulation of O₂ chemoreceptors induced elevations in fH and changes in net shunt. These tests attest that diverse populations of baroreceptors, O₂ chemoreceptors and temperature sensors have functional afferences generating autonomic reflexes even after the decerebration procedure. This indicates that the ANS of the decerebrated animals remains functional and capable of performing hemodynamic compensations to various perturbations. The study of the baroreflex through the pharmacological method indicated that the decerebrated animals maintain the gain and baroreflex limits similar to intact animals. We conclude that the decerebration doesn’t eliminate the reflexes generated by the centers of cardiovascular integration in the central nervous system (CNS) of rattlesnakes, keeping it functional like intact animals. In addition, isoflurane anesthesia suppresses the baroreflex of the decerebrated animals, modify the responses to autonomic function protocols, and causes a decrease of the mean arterial pressure. Thus, the use of anesthesia in the course of experimental protocols involving autonomic reflexes is not indicated.

Muitas vezes a compreensão dos mecanismos regulados pelo SNA, padrões de resposta, interação entre reflexos, dentre diversos outros fatores, requer estudos in vivo. O uso de anestésicos durante o curso dos protocolos experimentais e coleta de dados tem sido uma maneira de resolver os problemas éticos relacionados ao grau de instrumentação necessário para tais estudos. Os anestésicos, no entanto, deprimem o sistema nervoso e comprometem a conclusão de investigações que dependem do SNA funcional. Dessa maneira, nesse trabalho, buscamos validar uma preparação descerebrada com indivíduos da espécie Crotalus durissus. A descerebração consiste na inviabilização de áreas encefálicas superiores a fim de manter animais incapazes de processamento superior de informações e inviabilizando percepção e sensações de quaisquer tipos, incluindo estresse ou dor. Nestes animais, áreas do tronco encefálico e parte do hipotálamo, que participam da homeostase cardiovascular, são mantidas. No presente estudo, realizamos séries de testes fisiológicos para validação da presença de respostas autonômicas no modelo descerebrado. Além disso, realizamos o estudo do barorreflexo para compreensão de quão efetivo é o modelo em representar o animal intacto. Os animais descerebrados foram capazes de manter as variáveis hemodinâmicas constantes durante o protocolo de levantamento da cauda. Na manobra de levantamento da cabeça a pressão arterial média sistêmica (PAM sis) se elevou para garantir o suprimento adequado de sangue na cabeça. Além disso, durante aplicação de calor superficial, a frequência cardíaca (fH) se elevou, uma resposta cardiovascular típica de membros do grupo Squamata durante o aquecimento. A estimulação farmacológica de quimiorreceptores de O₂ através de injeções de cianeto de sódio produziu elevações na (fH) e alterações do net shunt. Estes testes atestam que diversas populações de barorreceptores, quimiorreceptores de O₂ e sensores de temperatura superficial possuem aferências funcionais gerando reflexos autonômicos esperados mesmo após o procedimento de descerebração. Isso indica que o SNA dos animais descerebrados se mantém funcional e capaz de executar compensações hemodinâmicas à diversas perturbações. O estudo do barorreflexo foi realizado em animais descerebrados e intactos. Através do método farmacológico, observamos que os animais descerebrados mantêm o ganho e os limites do barorreflexo similar à animais intactos. Com isso concluímos que a descerebração não elimina os reflexos gerados pelos centros de integração cardiovascular no sistema nervoso central (SNC) das cascavéis, mantendo-o com funcionamento equivalente ao animal intacto, mesmo sem as interações superiores. Além disso, a anestesia com isoflurano suprime as respostas barorreflexas dos animais descerebrados e altera as respostas aos protocolos de função autonômica, além de causar diminuição da Pa média dos animais. However, anesthetics depress the nervous system and compromise the conclusions from investigations that rely on functional ANS. Thus, in this work, we sought to validate a decerebrated preparation with individuals of the species Crotalus durissus. Decerebration consists in the ablation of superior brain areas in order to maintain preparations that are incapable of superior processing, perception and sensations of any type, including stress or pain. In these preparations, areas of the brainstem and part of the hypothalamus are maintained. Thus, the cardiovascular homeostasis is maintained. In the present study, we performed series of physiological tests for validated the presence of autonomic responses in the model. In addition, we performed a baroreflex study to understand how effective is the model in representing the intact animal. Decerebrated animals were able to keep hemodynamic variables constant during the tail lift protocol. In the head lift maneuver, the systemic mean arterial pressure (MAP sys) increased to ensure adequate blood supply to the head. In addition, during surface heat application, the heart rate (fH) increased, a cardiovascular response typical of Squamata group members during heating. Pharmacological stimulation of O₂ chemoreceptors induced elevations in fH and changes in net shunt. These tests attest that diverse populations of baroreceptors, O₂ chemoreceptors and temperature sensors have functional afferences generating autonomic reflexes even after the decerebration procedure. This indicates that the ANS of the decerebrated animals remains functional and capable of performing hemodynamic compensations to various perturbations. The study of the baroreflex through the pharmacological method indicated that the decerebrated animals maintain the gain and baroreflex limits similar to intact animals. We conclude that the decerebration doesn’t eliminate the reflexes generated by the centers of cardiovascular integration in the central nervous system (CNS) of rattlesnakes, keeping it functional like intact animals. In addition, isoflurane anesthesia suppresses the baroreflex of the decerebrated animals, modify the responses to autonomic function protocols, and causes a decrease of the mean arterial pressure. Thus, the use of anesthesia in the course of experimental protocols involving autonomic reflexes is not indicated.
