Níveis da secretase ADAM10 em idosos com diabetes mellitus tipo 2 e doença de Alzheimer

Resumo: | Alzheimer's disease (AD) is a gradual and progressive neurodegenerative disease characterized by predominant cortical atrophy in the medial temporal lobe and microscopically extensive neuronal losses and deposits called neurofibrillary tangles and senile plaques. From the results of the Rotterdam study in 1992, it is known that type 2 diabetes mellitus (T2DM) increases the risk of dementia. Patients with T2DM have structural brain changes and cognitive impairment, with a high risk of developing AD. These data suggest that insulin may play a key role in brain activity and memory formation. Our group has been studying peripheral AD biomarkers since 2010. Our results indicate that ADAM10 (A Disintegrin And Metalloprotease), the α-secretase that inhibits the formation of senile plaques and, therefore, is protective against AD, is active and decreased on platelets. Conversely, ADAM10 is inactive and increased in older adults with this dementia than cognitively healthy older adults. In this sense, this study aimed to verify the plasma ADAM10 levels among cognitively healthy older adults without glycemic alteration, older adults with AD, older adults with T2DM and older adults with the concomitant presence of AD and T2DM. The results showed a significant difference between plasma ADAM10 levels between all groups compared to older adults with AD+T2DM. Therefore, alterations in the insulin pathway can cause changes in ADAM10 levels and contribute to the worsening of AD. |

Therefore, alterations in the insulin pathway can cause changes in ADAM10 levels and contribute to the worsening of AD.A doença de Alzheimer (DA) é uma doença neurodegenerativa gradual e progressiva, caracterizada microscopicamente pela formação de depósitos denominados placas senis e emaranhados neurofibrilares, que geram toxicidade e colapso neural, resultando em perdas neuronais extensas e atrofia cortical predominante no lobo temporal medial. Desde os resultados do estudo de Rotterdam em 1992, sabe-se que o diabetes mellitus tipo 2 (DM2) aumenta o risco de demência. Pacientes com DM2 apresentam alterações cerebrais estruturais e comprometimento cognitivo, com elevado risco de desenvolver DA. Estes dados sugerem que a insulina pode desempenhar um papel fundamental com relação a atividade cerebral e com a formação da memória. Nosso grupo vem estudando biomarcadores periféricos para a DA desde 2010. Nossos resultados indicam que a ADAM10 (A Disintegrin And Metalloprotease), a α secretase que inibe a formação das placas senis e, portanto, é protetora contra a DA, está ativa e tem seus níveis diminuídos em plaquetas. De modo contrário, a ADAM10 está inativa e com níveis aumentados no plasma de idosos com esta demência, quando comparado a idosos cognitivamente saudáveis. Neste sentido, o objetivo deste estudo foi verificar os níveis da ADAM10 no plasma de idosos cognitivamente saudáveis e sem alteração dos níveis glicêmicos, de idosos com DA, de idosos com DM2 e de idosos com a presença concomitante de DA e DM2. Os resultados mostraram diferença significativa entre os níveis de ADAM10 plasmática entre todos os grupos quando comparado ao grupo de idosos com DA+DM2.
