Caracterização de rizobactérias associadas à Solanum lycocarpum St. Hil. quanto ao potencial biotecnológico para promoção de crescimento vegetal, enzimas e agentes antimicrobianos

Resumo: | Solanum lycocarpum St. Hil., endemic plant of the Brazilian tropical savanna, known as the “Cerrado”, has been studied for its medicinal properties and for having the ability to grow in acidic and nutrient-poor lands, attracting interest in the investigation of its rhizospheric microbiota, which includes plant growth-promoting rhizobacteria (PGPR). Industrial enzymes and antibiotics can also be obtained from them. In this work, the biological nitrogen fixation capacity was tested in NFb semi-solid culture medium and 32.6% of the isolates showing positive results. In the inorganic phosphate solubilization tests, 45.8% of the isolates demonstrated this capacity in nutrient agar supplemented with insoluble phosphate. The solubilization index (SI) for calcium phosphate by the bacterial isolates grown in solid medium varied from 0 to 4.833. All bacteria produced indole acetic acid (ranging from 0.07 μg.mL-1 to 134.1 μg.mL-1) in Tryptic Soy Broth (TSB) 10% + L-tryptophan medium. In assays against pathogens, antagonistic activity was demonstrated against a spectrum of fourteen phytopathogenic fungi through the technique of direct pairing in Potato Dextrose Agar (PDA) medium and three clinical pathogens using the agar block method. The bacterial isolates were striated in special mediums whose carbon sources were substrates of the enzymes in the enzyme activity assays and 88.4% of them had at least one of these activities. The best isolates were identified by partial sequencing of the 16S gene as belonging to the genera Bacillus, Burkholderia and Microbacterium. |

A Solanum lycocarpum St. Hil., conhecida popularmente como lobeira, é uma planta característica deste bioma, que têm sido estudada pela capacidade de se desenvolver em terras ácidas e pobres em nutrientes; ademais, possui propriedades medicinais descritas em literatura, tornando interessante o estudo de sua microbiota rizosférica também no contexto da resistência microbiana, problema preocupante para a saúde mundial que aumenta a severidade das doenças infecciosas e os custos de tratamento. O presente trabalho teve como objetivos a caracterização bioquímica de cento e trinta e uma bactérias associadas à rizosfera de lobeira avaliando, in vitro, o potencial desses micro-organismos para a fixação biológica de nitrogênio (FBN), solubilização de fosfato (SF), produção de ácido indol-acético (AIA) e controle biológico por meio de antagonismo à fungos fitopatógeno; adicionalmente, foram avaliados o perfil de atividade enzimática e o potencial de produção de antibióticos pelos isolados. A identificação dos melhores isolados também foi realizada. Quanto aos testes in vitro, a FBN foi testada em meio de cultura semi-sólido livre de nitrogênio, a SF foi testada em meio ágar nutriente suplementado com fosfato insolúvel e o AIA foi testado o meio caldo Triptona de Soja (TSB) 10% + L-triptofano. No teste de antagonismo, houve uma seleção massal utilizando discos com micélios de 14 fungos fitopatógeno que foram depositados em placas contendo meio Potato Dextrose Agar (PDA) juntamente com os isolados e as bactérias com melhores resultados foram selecionadas para o teste de pareamento direto com os mesmos fitopatógenos. Para os ensaios de atividade enzimática, os isolados foram estriados em meios especiais cujas fontes de carbono eram substratos das enzimas de interesse. A antibiose contra três diferentes patógenos clínicos foi avaliada pelo método do bloco de gelose. Na identificação molecular foi utilizado o gene 16S e as sequências foram comparadas no software MEGA X. Do total, 32,6% dos isolados obtidos apresentaram resultados positivos para fixação biológica de nitrogênio atmosférico, todos produziram AIA (variando de 0,07 μg.mL-1 a 134,1 μg.mL-1) e 45,8% solubilizaram fosfato inorgânico. Os testes de atividade enzimática in vitro revelaram que 42,9% dos isolados endofíticos apresentaram atividade amilolítica, 42,9% celulolítica, 47,3% esterolítica, 27,7% lipolítica, 31,3% pectinolítica e 60,7% proteolítica. Nos ensaios contra patógenos, foi demonstrado atividade antagônica contra um espectro de 14 fungos fitopatogênicos e 3 patógenos de importância clínica.
