PLANT EXTRACTS (Nicotiana glauca and Chrysanthemum vulgaris) IN A VACCINE TO BOVINE HERPESVIRUS TYPE 5 (BHV-5)

Resumo: | The effect of Nicotiana glauca and Chrysanthemum vulgaris leave extracts on the humoral response in cattle vaccinated with an inactivated vaccine to bovine Herpesvirus type 5 (BHV-5) were examined. Two inactivated oil-adjuvanted BHV-5 vaccines were prepared with an infectious titre (pre-inactivation) of 10 84DICC/ ml, inactivated with binary ethylenimine (BEI) and emulsified in oil adjuvant. Nicotiana glauca and Chrysanthemum vulgaris extracts were added . (5mg of each extract per dose) as a coadjuvant to one of the vaccines (vaccine B). Two groups of nineteen calves were vaccinated with 3 doses (3 ml) at four week intervals between doses. Neutralizing antibody leveis in sera were determined at different time intervals after vaccination. Thirty days after the administration of the third dose the mean neutralizing antibody titre was 96 for vaccine A and 80 for vaccine B. There was no significant difference between the antibody titres induced by the two vaccines (p>0.05). These results indicate that extracts of these plants had no adjuvant effect on the inactivated BHV-5 vaccine . |

Este trabalho analisa o efeito da adição do extrato das folhas de Nicotiana glauca e Chrysanthemum vulgaris em vacina inativada contra Herpesvírus bovino tipo 5 (BHV-5) sobre a resposta imune humoral em bovinos vacinados. Duas vacinas para BHV-5 inativadas com etileno mina binária e emulsificadas com adjuvante oleoso foram produzidas a partir de uma suspensão viral com título de 10 8 . 6 DICC50/ml. Uma das vacinas (vacina 13) teve a incorporação de extrato bruto das plantas Nicotiana glauca e Chrysanthemum vulgaris na dose de 5mg de extrato de cada planta por dose. As vacinas foram testadas em dois grupos de 19 bovinos, vacinados com 3 doses (3m1) de cada vacina com intervalo mensal. Os títulos de anticorpos neutralizantes foram determinados em diferentes intervalos após a vacinação através do teste de soroneutralização. Trinta dias após a terceira dose vacinal a vacina A induziu título médio de anticorpos de 96 e a vacina B de 80. Na análise estatística não houve diferença significativa entre os títulos de anticorpos induzidos pelas duas vacinas (p>0,05). Os resultados indicaram que a adição dos extratos destas plantas não influenciou a resposta imune humoral nesta vacina. Departamento de Diagnóstico e Pesquisa Agropecuária.
