Resumo: | Introduction: Currently diabetes mellitus (DM) is increasing, standing out as a worldwide public health problem, causing negative implications to various dimensions that make up the community settings. Thus, a special attention with interdisciplinary view has been employed at different levels and prevention of diabetes coping. In this way, several plant species have been used to treat or combat the symptoms of diabetes mellitus, both in animal models and clinical studies/population studies in humans. Objective: The aim of this study was to evaluate the effects of subchronic ingestion of different doses of methanol extract of plant Baccharis dracunculifolia in an animal model of DM on the biochemical, hormonal and histological parameters. Methods: They were used in this studt 56 Wistar adults male rats was treated for 28 days. The animals were divided into 8 groups, with 07 animals per group: CT - Tween Control ("Tween 80" + distilled water), CB - Control Baccharis (Extract B dracunculifolia - 200 mg / kg), CDT - Diabetic Control ("Tween 80"+ distilled water) DS - Sulfonylurea Diabetics (Sulfonylurea - 10 mg / kg), DB50 - Diabetic Baccharis 50 mg (Extract B dracunculifolia - 50 mg / kg), DB100 - Diabetic Baccharis (100 mg Extract B. dracunculifolia - 100 mg / kg), DB200 - Diabetic Baccharis (200 mg Extract B. dracunculifolia 200 mg / kg) DB200S - 200 mg Baccharis Diabetics + Sulfonylurea (Extract B dracunculifolia - 200 mg / kg + Sulfonylurea - 10 mg / kg). The diabetes induction was performed by administration of 60 mg / kg of streptozotocin (STZ).The animals were evaluated for weight, feed intake, water intake and fasting blood glucose and postprandial (OGTT). At the end of subchronic treatment, the animals were euthanized, and blood and tissues samples were collected for determination of biochemical and hormonal parameters, and preparation of histological slides. Results and Discussion: There was statistical difference for DB50 and DB100 groups regarding the CDT group to creatinine parameters (p <0.05), urea (p <0.05), triglycerides (p <0.05) and plasma glucose (p <0.05). The plasma insulin levels was statistically significant (p <0.01) between the evaluated diabetic groups, an increase of 81,1% to 94,4 % between the three doses of pure extract ( DB50 , and DB100 DB 200) , compared to diabetic untreated group. In fasting glucose and postprandial , the DB50 groups , DB100 , DB200 and DB200S, showed a decrease of 54,3 % to 63,4 % to fasting and 37,9 % to 45,0 % in the postprandial the end monitoring. All treatments with B. dracunculifolia, especially the groups that received the extract exclusively plant (DB50, DB100 and DB200), were effective in minimizing tissue damage of diabetes installed throughout the experiment histological qualitative assessment. Conclusion: Methanol extract of Baccharis dracunculifolia decresed levels of fasting capillary glucose and postprandial in rats with diabetes induced by STZ in different doses. There was a decrease in plasma glucose, triglycerides, creatinine and urea levels, in STZ-induced animals treated with the plant, particularly at concentrations of 50 and 100 mg. The Baccharis dracunculifolia extract presented an apparent damage protection in organs (liver, kidney and pancreas) in induced diabetic rats. The groups treated with the extract of B. dracunculifolia have higher concentrations of plasma insulin release in all administered doses of the extract. |

Deste modo, uma atenção especial de forma interdisciplinar tem sido empregada aos diferentes níveis preventivos e de enfretamento do diabetes. Nesse sentido, várias espécies de plantas vêm sendo utilizadas para tratar e ou combater os sintomas do diabetes mellitus, tanto em modelos animais como estudos clínicos ou populacionais em seres humanos. Objetivo: Avaliar os efeitos da ingestão subcrônica em diferentes doses do extrato metanólico da planta Baccharis dracunculifolia, em um modelo animal de DM sobre os parâmetros bioquímicos, hormonais e histológicos. Métodos: Foram utilizados 56 ratos machos adultos da linhagem Wistar tratados por 28 dias. Os animais foram organizados em 8 grupos de 07 indivíduos da seguinte forma: CT - Controle Tween ("Tween 80" + Água destilada ), CB - Controle Baccharis (Extrato B. dracunculifolia - 200 mg/kg), CDT - Controle Diabéticos ("Tween 80" + Água destilada), DS - Diabéticos Sulfoniluréia (Sulfoniluréia - 10 mg/kg), DB50 - Diabéticos Baccharis 50 mg (Extrato B. dracunculifolia - 50 mg/kg), DB100 - Diabéticos Baccharis 100 mg (Extrato B. dracunculifolia - 100 mg/kg), DB200 - Diabéticos Baccharis 200 mg (Extrato B. dracunculifolia - 200 mg/kg), DB200S - Diabéticos Baccharis 200 mg + Sulfoniluréia (Extrato B. dracunculifolia - 200 mg/kg + Sulfoniluréia - 10 mg/kg). A indução do diabetes foi realizada pela administração de 60 mg/kg de estreptozotocina (STZ). Os animais foram avaliados quanto a evolução ponderal, consumo alimentar, ingestão hídrica e glicemia capilar e pós-prandial (TOTG). Ao final do tratamento subcrônico os animais foram eutanasiados para as coletas de sangue e tecidos para determinação de parâmetros bioquímicos, hormonais e confecção de lâminas histológicas. Resultados e Discussão: Para os grupos DB50 e DB100 em relação ao grupo CDT houve redução nos níveis de creatinina (p<0.05), uréia (p<0.05), triglicérides (p<0.05) e glicose plasmática (p<0.05). Os níveis de insulina plasmática apresentaram significância estatística (p<0.01) entre os grupos diabéticos, com elevação de 81,1% a 94,4% entre as três doses administradas de extrato puro (DB50, DB100 e DB 200), quando comparados ao grupo diabético não tratado. Na glicemia capilar em jejum e pós-prandial, os grupos DB50, DB100, DB200 e DB200S, apresentaram diminuição de 54,3% a 63,4% ao jejum e de 37,9% a 45,0% no pós-prandial ao final do acompanhamento. Todos os tratamentos com B. dracunculifolia, em especial os grupos que receberam o extrato da planta de forma exclusiva (DB50, DB100 e DB200), foram eficazes em minimizar os danos teciduais do diabetes instalado ao longo do experimento na avaliação qualitativa histológica. Conclusão: O extrato metanólico de Baccharis dracunculifolia promoveu queda dos níveis de glicose capilar ao jejum e pós-prandial de ratos induzidos à diabetes por STZ nas diferentes doses administradas. Houve redução de valores de glicose plasmática, triglicerídeos, creatinina e uréia de animais STZ induzidos tratados com a planta, especialmente nas concentrações de 50 e 100 mg. O extrato de B. dracunculifolia apresentou aparente proteção de danos em órgãos (fígado, rins e pâncreas) de animais diabéticos induzidos.
