Valor genético para abelhas africanizadas selecionadas para produção de geleia real com marcadores moleculares

Resumo: | The aim of this research was to evaluate the genetic value for royal jelly production in Apis mellifera L. Africanized with the compilation of data collected since 2006. The genetic informations of selected and evaluated colonies were obtained using techniques of full DNA extraction from nursing worker bee's thorax, with molecular markers for MRJP3 protein, which is well characterized in Apis mellifera L. For larvae transfer to produce queens and royal jelly a heated room was used, with temperature between 30ºC and 35ºC and air humidity above 60%. The larvae aged between 12 and 36h were transferred from its original cell to acrylic cups. The royal jelly collection was done after 66 to 72h with suction apparatus. From colonies information and genealogical structure, genetic values were predicted for colonies and queens considering the parameters: acceptance (%), total royal jelly (mg) and royal jelly per cup (mg). The Bayesian inference was used with the software Multiple Trait Gibbs Sampling in Animal Models, Gibbs chains with 58500 cycles, resulting in 650.000 cycles with disposal and removal intervals of 65000 and 10, respectively. The predicted genetic values from colonies were classified into superior and inferior. Comparing the genetic values averages as a function of genotypes, the test of multiple average comparisons were realized, implemented in routine PROC GENMOD of the Statistical Analysis System. Using the same routine the classification probability was estimated in superior and inferior categories of these genotypes, considering binomial distribution of data with log-link function. The classification probabilities for each genotype were tested by the T test at 5% significance. The environmental effects of year, time and type of colony were considered as having flat distribution and collection as qui-squared distribution. The results shown an increase of alleles C, D and E in matrices honeybee colonies in which, the alleles D and E (relating to MRJPs) had the most valuable genes for royal jelly production. The alleles D, E and C are the most important when evaluating the parameters of production, acceptance, total royal jelly and royal jelly per cup, and promptly, the genotype DE had emphasis on royal jelly production. The genotypes DE, DC and CE should be maintained for royal jelly production, and other genotypes should be discarded because they had the worst performances. |

As informações genéticas das colônias selecionadas e avaliadas foram obtidas utilizando técnicas de extração do DNA total do tórax de abelhas operárias nutrizes com marcadores moleculares para proteína MRJP3, bem caracterizada em Apis mellifera L. Para as transferências de larvas na produção de rainhas e geleia real foi utilizada sala climatizada com temperaturas médias entre 30ºC e 35ºC e umidade relativa do ar acima de 60%. As larvas em idade entre 12 e 36h foram transferidas de sua célula de origem para cúpulas de acrílico. A coleta de geleia real foi feita de 66 à 72h com aparelho de sucção. A partir das informações das colônias e da estrutura genealógica foram preditos os valores genéticos das colônias e rainhas para as características aceitação de larvas transferidas - %, geleia real total - mg e geleia real por cúpula - mg. Foi utilizado o modelo animal com inferência bayesiana do software Multiple Trait Gibbs Sampling in Animal Models, cadeias de Gibbs de 58500 ciclos, resultantes de 650.000 ciclos, com intervalos de descarte e retirada de 65000 e 10, respectivamente. A partir dos valores genéticos preditos, as colônias foram classificadas em superior e inferior. Para comparar as médias dos valores genéticos em função dos genótipos, procedeu-se o teste de comparação múltipla de médias, implementado na rotina PROC GENMOD do Statistical Analysis System. Utilizando a mesma rotina foi estimada a probabilidade de classificação nas categorias superior e inferior desses genótipos considerando a distribuição de dados binomial com função de ligação logarítmica. As probabilidades de classificação para cada genótipo foram testadas por meio do teste T em nível de 5% de significância. Foram considerados os efeitos ambientais de ano, época e modelo de colônia (padrão Langstroth) como apresentando distribuição plana, e de coleta como distribuição de qui-quadrado. As pesquisas mostraram um aumento dos alelos C, D e E nas abelhas matrizes que fazem parte deste sistema de avaliações quantitativas e moleculares na produção de geleia real, sendo os alelos D e E - referentes às MRJP`s - encontrados no grupo de maior valor genético para produção. Os alelos D, E e C são os mais importantes quando avaliamos as características de aceitação, geleia real total e por cúpula, e que pontualmente, foi o genótipo DE que se destacou na produção de geleia real.
