Alterações no tecido intestinal e o envolvimento de bactérias na fase crônica da infecção de Trichuris muris em camundongos

Resumo: | The parasitism caused by Trichuris spp. Has global relevance, impacting the medical and veterinary area. The T. trichiura species infects about 795 million people, being predominant in countries with tropical climate that present precarious conditions of basic sanitation. Human infection mainly affects children in the school age group, causing impairment in their physical and cognitive development. The tricurid nematodes develop a strategy of fixation in the host that promotes perforations in epithelial mucosa and to understand the parasite/host interaction is necessary to better describe the infection establishment process, the development of the parasite and the pathophysiological evolution of this parasite. By expanding this information, the developments may be linked to the control and the search for new chemotherapeutic alternatives. In the present work we used the T. muris experimental infection model in Swiss webster mice to evaluate the changes promoted by parasitism in the chronic phase, including histopathological analyzes of the large intestine, biochemical and hematological parameters, composition and alteration in the intestinal microbiota of the mice and the analyses of the bacteria in the submucosa of the cecum. The infection developed in the animals an iron deficiency anemia, due to a strong iron deficiency, the thickening of the mucosa and submucosa due to an intense cellular infiltrate. In addition, we identified the presence of bacteria in the inner layers of the intestinal tissue. The microorganisms can be accessed the submucosa by mucosal ruptures promoted by nematode during the infection. We proposed that the tissue alterations and the intense cellular infiltrate was associated of the nematode presence and the bacteria invasion. The understanding of the bacterial invasion mechanisms and the interaction of these microorganisms with the nematode opens new frontiers in relation to the pathophysiological mechanisms of the human and veterinary trichuriasis. |

A espécie T. trichiura infecta cerca de 795 milhões de pessoas, sendo predominante em países com clima tropical que apresentam condições precárias de saneamento básico. A infecção humana afeta principalmente crianças na faixa etária escolar, causando prejuízo no seu desenvolvimento físico e cognitivo. Os nematoides tricurídeos desenvolvem uma estratégia de fixação no hospedeiro que promove perfurações na mucosa intestinal e compreender a biologia desta interação parasito/hospedeiro é necessário para melhor descrever o processo de estabelecimento da infecção, o desenvolvimento do parasito e a evolução fisiopatológica desta parasitose. Ampliando estas informações, os desdobramentos podem estar atrelados ao controle e a busca de novas alternativas quimioterápicas. No presente trabalho utilizamos o modelo de infecção experimental T. muris em camundongos suíços para avaliar as alterações promovidas pelo parasitismo em fase crônica, incluindo análises histopatológicas do intestino grosso, parâmetros bioquímicos e hematológicos, composição e alteração na microbiota intestinal dos camundongos e a análise de estruturas bacterianas na submucosa do ceco. A infecção desenvolveu nos animais uma anemia ferropriva, devido a forte carência de ferro, o espessamento da mucosa e submucosa em decorrência de um intenso infiltrado celular. Além disso, identificamos a presença de bactérias nas camadas mais internas do tecido intestinal. Estes micro-organismos possivelmente tiveram acesso à estas camadas por rupturas na mucosa epitelial do tecido intestino grosso, promovida pelo nematoide durante o desenvolvimento da infecção. Estamos propondo que as alterações teciduais e o intenso infiltrado celular observado está associado à presença do parasito e das bactérias invasoras.
