Resumo: | Invasion of bacteria capable of causing cystitis and / or pyelonephritis characterizes UTI. Escherichia coli is the main bacterial species associated with this infection, due to its importance in causing UTI, and some of this species are called UPEC (Uropathogenic Escherichia coli). In the present study, 11 UPEC strains isolated from urine of patients from the HUPE nephrology clinic with symptomatic or asymptomatic infections were studied. The presence of 5 virulence factors was investigated through simple and multiplex PCR techniques to verify the genes encoding adhesins P (pap), fímbria S (sfa), sideroforo (aerobactinin) and alpha-hemolysins (hly); islands of pathogenicity (virulence) through the PAI marker. The antimicrobial resistance profiles and presence of the main resistance genes such as blaNDM-1, blaVIM-1, blaIMP-1, blaKPC (genes encoding the production of metalobetalactamase enzymes) were evaluated; blaCTX (encoding ESBL) and oxa-48 gene (which encodes the production of OXA-carbapenemase). To investigate the pathogenesis of these infections, we used the adhesion and invasion tests in VERO cells where we had 100% adherence and invasion, as well as the use of scanning electron microscopy (SEM), as a methodological alternative for the registration of adhesion and invasion of E in VERO cells. The strains of UPECs tested did not present virulence factors, or had at least one of these factors. We used to characterize the clonal relationship of the strains studied in the study of pulsed field electrophoresis (PFGE) and the phylogenetic relationship by determining the groups A, B1, B2 and D of these microorganisms, which presented as a prevalence of groups D followed by B2. The joint analysis of these data contributed to determine, together with the formation of biofilm and the resistance, that there is no relationship between each other, all these analyzed parameters |

A Escherichia coli é a principal espécie bacteriana associada a esta infecção, devido a sua importância em causar ITU, e algumas dessa espécie receberem a denominação de UPEC (Escherichia coli uropatogênica). No presente trabalho foram estudadas 11 cepas de UPEC isoladas de urina de pacientes do ambulatório de nefrologia do HUPE, com infecções sintomática ou assintomática. Foi pesquisada a presença de 5 fatores de virulência, através das técnicas de PCR simples e multiplex para verificação dos genes que codificam adesinas P (pap), fímbria S (sfa), sideroforo (aerobactina- aer) e alfa-hemolisinas (hly); ilhas de patogenicidade (virulência) através do marcador PAI. Foram avaliados os perfis de resistência a antimicrobianos e a presença dos principais genes de resistência como: blaNDM-1, blaVIM-1, blaIMP-1, blaKPC (genes que codificam a produção de enzimas metalobetalactamase); blaCTX (que codifica a produção de enzimas betalactamase de espectro ampliado - ESBL) e o gene oxa-48 (que codifica a produção de OXA-carbapenemase). Para investigarmos a patogênese dessas infecções usamos os testes de aderência e invasão em células VERO onde tivemos 100% de aderência e invasão, assim como a utilização da microscopia eletrônica de varredura (MEV), como alternativa metodológica para o registro de aderência e invasão de E. coli em células VERO. As cepas de UPECs testadas não apresentaram fatores de virulência, ou apresentaram pelo menos um desses fatores. Utilizamos para caracterizar a relação clonal das cepas em estudo a eletroforese de campo pulsado (PFGE) e a relação filogenética pela determinação dos grupos A, B1, B2 e D destes microrganismos, que apresentaram como prevalência de grupos D seguido de B2.
