Fumaça de cigarro durante a adolescência de camundongos fêmeas e machos: efeitos da exposição e da abstinência sobre a adiposidade

Resumo: | Tobacco use is one of the great pandemics of all time. The smoking rate among male adolescents, and especially female growing. So the objective was to evaluate the influence of smoking on puberty and on abstinence in adult life, on issues related to body composition and adiposity Swiss mice, females and males. At 35 days of age, females and males were subjected to the smoke chamber, where they were divided into two experimental groups. After fifteen days of exposure, half the animals in each group were sacrificed. The other half remained abstinent for thirty days. At the end of exposure to smoke or after thirty days of abstinence, the animals were evaluated in vivo for determination of glucose tolerance and determination of body composition by DXA and then were sacrificed to collect tissue and conducting other analyzes. During exposure to smoke, the body mass of animals-smoking males and females (F / 15) was lower than that of their respective controls, with low or no weight gain during the 15 d. During this period, the daily caloric intake and food intake were similar between C and F / 15 groups. During the 30 days of abstinence, the trend to ↑ food intake determined higher caloric intake in ex-smoker group, which was translated by an increase in body mass in females but not in males. Exposure to smoke for females was a factor inducing the gain of fat trunk and total reflected by the mass gain of ART. These data are confirmed by the higher fat content and leptin levels, checked in smoking females. The glycemic index rating, females exposed to smoke had increased plasma concentrations of glucose and insulin, however, with ↓ activity of pancreatic beta cells, indicating a possible change in insulin action. The ART was also assessed in relation to oxidative stress, where catalase showed no alteration in females but ↑ in males, GPx activity was at its ↓ in females and males. The protein oxidation was determined by carbonylation which showed an increase in females. The cessation of smoking, does not reverse the damage caused by smoke, whose toxic potential remains significant, affecting body composition, structure and function of the intra-abdominal adipose tissue. |

Os índices de tabagismo entre adolescentes do sexo masculino e, principalmente, do sexo feminino vêm crescendo. Assim o objetivo foi avaliar a influência do fumo na puberdade e na abstinência na vida adulta, sobre aspectos relacionados à composição corporal e à adiposidade de camundongos Swiss, fêmeas e machos. Aos 35 dias de idade, as fêmeas e os machos foram submetidos à câmara de fumaça, onde foram divididos em dois grupos experimentais. Após quinze dias de exposição, metade dos animais de cada grupo foi sacrificada. A outra metade permaneceu em abstinência por trinta dias. Ao final da exposição à fumaça ou após trinta dias de abstinência, os animais foram avaliados in vivo, para determinação da tolerância a glicose e para determinação da composição corporal por DXA e, em seguida foram sacrificados para coleta de tecidos e realização das demais análises. Durante a exposição à fumaça, a massa corporal dos animais fumantes fêmeas e machos (F/15) foi menor que a de seus respectivos controles, com baixo ou nenhum ganho de massa durante os 15 d. Nesse período, a ingestão alimentar diária e a ingestão calórica foram semelhantes entre os grupos C e F/15. Durante os 30 dias de abstinência, a tendência ao ↑ de ingestão alimentar determinou maior ingestão calórica no grupo ex-fumante, que foi traduzida pelo aumento da massa corporal nas fêmeas, mas não nos machos. A exposição à fumaça para as fêmeas foi um fator indutor do ganho de gordura do tronco e total, refletida pelo ganho de massa de TAR. Esses dados são confirmados pelo maior índice de adiposidade e concentração plasmática de leptina, verificados nas fêmeas fumantes. Na avaliação de índice glicêmico, fêmeas expostas à fumaça apresentaram concentrações plasmáticas aumentadas de glicose e insulina, no entanto, com ↓ atividade das células beta pancreáticas, indicando possível alteração da ação da insulina. O TAR também foi avaliado em relação ao estresse oxidativo, onde a catalase não revelou alteração nas fêmeas mas ↑ nos machos e a GPx, sua atividade se encontrava ↓ nas fêmeas e ↑ nos machos. A oxidação proteica foi determinada por carbonilação que evidenciou aumento nas fêmeas.
