Influência do hormônio do crescimento nos processos celulares e moleculares da fase inflamatória do reparo tecidual cutâneo

Resumo: | In the present study, we investigated in vivo and in vitro the effects of growth hormone (GH) on cellular and molecular processes involved in the inflammatory phase of cutaneous tissue repair in mice. For this purpose, we used 6-month-old male mice of Swiss strain (CEUA / UFAL No. 51/2014). The induction of the wound was performed, after anesthetic administration, with the aid of a 1cm diameter metal ring. Then, the animals were treated with GH (10−7M) for 3 consecutive days, once a day. The control group received only vehicle. The skin fragments were stained with HE for the count of inflammatory cells. Injured skin samples were performed for quantification of cytokines (IL-6, IL-10, TNF-a) involved in inflammatory cellular responses by ELISA. Immunohistochemistry assay was used to evaluate the presence of macrophages and endothelial cells, and the deposition of extracellular matrix molecules: collagen III, fibronectin and laminin. Liver and injured skin fragments were used to evaluate the IGF-1 mRNA expression. In vitro, J774 lineage macrophages and thymic endothelial cells (tEnd.1) were used to evaluate the formation of capillary-like structures in matrigel. Fibroblasts from the 3T3 strain were used for cell migration assay. Initially, the topical treatment with GH for 3 days hás accelerated the recruitment of macrophages and lymphocytes without altering IL-6 levels when compared to control. However, topical use of GH revealed increased levels of IL-10 and decreased levels of TNF-a. In vitro, GH treatment did not alter the IL-6 and IL-10 levels produced by macrophages treated with GH (10−7M). However, GH-treated macrophages conditioned medium stimulated endothelial cells in the formation of capillary-like structures in matrigel. In addition, GH increased the fibroblasts migration. It was further found that topical treatment with GH stimulated angiogenesis, type III collagen deposition of fibronectin and laminin. Finally, gene expression analysis revealed that topical treatment with GH does not alter mRNA levels for IGF-1 in both liver and skin. Taken together, our data suggest that GH accelerates the healing of cutaneous wounds, anticipating macrophage recruitment, increasing the deposition of ECM proteins and stimulating angiogenesis, independent of IGF-1. |

Para este propósito, foram utilizados camundongos, com 6 meses de idade, machos da linhagem Swiss (CEUA/UFAL No 51/2014). A indução da ferida foi realizada, após a administração de anestésico, com auxilio de um punch dermatológico de 1cm de diâmetro. Após estes procedimentos, os animais foram tratados topicamente com GH (10−7 M) por 3 dias consecutivos, uma vez ao dia. O grupo controle recebeu apenas veículo. Os fragmentos de pele foram submetidos a coloração por hematoxilina e eosina (HE) para a contagem de células inflamatorias. Amostras da pele injuriada foram usadas para quantificação das citocinas envolvidas em respostas celulares inflamatórias pelo ensaio de imunoabsorção enzimática (ELISA). Por imunohistoquímica foi avaliada a presença de macrófagos e células endoteliais, e a deposição de moléculas de matriz extracelular: colágeno III, fibronectina e laminina. Fragmentos do fígado e da pele que sofreu injuria foram utilizados para avaliar a expressão do RNAm para o fator-1 de crescimento semelhante a insulna (IGF-1). In vitro, foram utilizados macrófagos da linhagem J774 e células endoteliais tímicas (tEnd.1) para avaliar a formação de estruturas semelhantes a capilares, em matrigel. Fibroblastos da linhagem 3T3 foram utilizados para o ensaio de migração celular. Inicialmente, demonstrou-se que o tratamento tópico com GH por 3 dias acelerou o recrutamento de macrófagos e linfócitos sem alterar os níveis de interleucina-6 (IL-6) quando comparado ao controle. No entanto, o uso tópico do GH revelou aumento dos níveis de IL-10 e diminuição nos níveis do fator de necrose tumoral alfa (TNF-a). In vitro, o tratamento com GH não alterou os níveis de IL-6 e IL-10 produzidos pelos macrófagos, na concentração de 10−7 M, no entanto, o meio condicionado de macrófagos tratado com GH estimulou as células endoteliais na formação de estruturas semelhantes a capilares, em matrigel. Além disso, o GH aumentou a migração de fibroblastos. Verificou-se ainda que o tratamento tópico com GH estimulou a angiogênese a deposição de colágeno tipo III, a deposição de fibronectina e de laminina. Por fim, a análise da expressão gênica revelou que o tratamento tópico com GH não altera os níveis de RNAm para o IGF-1 no fígado e na pele.
