Ana Ligia ScottTese (doutorado) - Universidade Federal do ABC, Programa de Pós-Graduação em Ciência e Tecnologia/Química, Santo André, 2019.Neste trabalho foram analisadas as principais características estruturais da enzima dipeptidil peptidase-4 (DPP-4), de forma a compreender as características do sítio ativo, assim como as principais interações entre ligantes bioativos e o alvo biológico. Desta forma, este trabalho busca auxiliar o planejamento de novos inibidores, uma vez que esta enzima está relacionada com o controle dos níveis de glicose no organismo. Como o número de casos do diabetes mellitus tipo II vem aumentando em todo o mundo, este alvo tem grande relevância biológica e uma boa estratégia para auxiliar no tratamento é a utilização de fármacos que possuam inibição eficaz frente à DPP-4 e que apresentem efeitos colaterais mínimos. As técnicas usadas neste trabalho forneceram informações importantes para a fase de drug design, na qual foram consideradas as peculiaridades do alvo. Dentre as abordagens utilizadas, pode-se citar cálculo dos modos normais, detecção e caracterização de sítios de ligação, estudo das interações moleculares de inibidores, acoplamento molecular, estudos de QSAR 3D (CoMFA e CoMSIA), cálculos de propriedades ADMET, triagem virtual, teste enzimático e modificações moleculares por bioisosterismo. Com isto, foi possível detectar um 3º sítio de ligação da DPP-4 ainda não explorado em estudos relacionados como possível região de inibição desta enzima. Desta forma, a partir deste resultado foi possível avaliar as principais características deste sítio e requisitos moleculares que um ligante deve possuir para inibir eficazmente a enzima a partir desta região. Além disso, foram obtidos modelos de QSAR robustos e preditivos (CoMFA: q2= 0,863 e r2 = 0,991; e CoMSIA: q2 = 0,800 e r2 = 0,965). Estes modelos serviram de base para a proposta de modificações estruturais a partir da estratégia de isosterismo. Por fim, realizou-se triagem virtual em que os filtros utilizados no protocolo planejado especificamente para a enzima DPP-4 permitiram selecionar potenciais candidatos a inibidores da DPP-4. Um deles foi obtido comercialmente e testes biológicos frente à enzima foram realizados. Os resultados experimentais indicam que este composto apresenta atividade inibitória para DPP-4, mas necessita de otimização estrutural para melhoria de sua atividade, assim como testes futuros como citotoxicidade.In this study, the main structural characteristics of the enzyme known as dipeptidyl peptidase-4 (DPP4) were analyzed in order to understand the characteristics of the active site as well as the main interactions between bioactive ligands and the biological target. In this way, this study seeks to help the design of new inhibitors, since this enzyme is related to the control of glucose levels in the body. As the number of cases of type II diabetes mellitus is increasing worldwide, this target has great biological relevance and a good strategy to aid in the treatment is the use of drugs that promote effective inhibition of DPP-4 and minimum side effects. The techniques used in this study provided important information for the drug design, in which the peculiarities of the target were considered. The techniques used here include normal modes, detection and characterization of binding sites, study of molecular interactions of inhibitors, molecular docking, 3D QSAR studies (CoMFA and CoMSIA), calculation of ADMET properties, virtual screening, enzymatic assays and molecular modifications by bioisosterism. With this, it was possible to detect a 3rd DPP-4 binding site not exploited so far in related studies as a possible region for the inhibition of this enzyme. Thus, from this result it was possible to evaluate the main characteristics of this site and molecular requirements that a ligand must possess to effectively inhibit the enzyme from this region. In addition, robust and predictive QSAR models were obtained (CoMFA: q2 = 0.863 and r2 = 0.991; CoMSIA: q2 = 0.800 and r2 = 0.965). These models served as a basis for the proposal of structural modifications based on the strategy of isosterism. Finally, virtual screening was performed in which the filters used in the protocol specifically designed for the DPP-4 enzyme allowed to select potential candidates as DPP-4 inhibitors. One of them was commercially obtained and biological tests against the enzyme were performed.
