Resumo: | The present study was carried out to evaluate the effect of biochemical composition of seminal plasma (SP) on spermatozoa from caudal epididymis and ejaculated goat sperm, diluted in egg yolk-citrate (EYC) and egg yolk-tris (EYT). The ejaculates of 8 animals were collected in the morning and fresh semen was divided in order to obtain 4 treatments: EYC (washed ad no washed), EYT (washed ad no washed). On the same day, in the afternoon, the animals were orchidectomized to obtain spermatozoa from caudal epididymis, of which four aliquots of 100 L were removed. Two of them were added 120 L of autologous seminal plasma, consisted in other 4 treatments: EYC (spermatozoa from caudal epididymis with and without seminal plasma) e EYT (spermatozoa from caudal epididymis with and without seminal plasma). The seminal plasma was evaluated for biochemical composition. Samples of 8 treatments were analyzed for vigor, motility and sperm morphology, at 0 hour (fresh) and after 2 and 24 hours in storage at 5ÂC. Data were analyzed with the statistic software SAS@. The following results were found. In grouping by the concentration of fructose and chlorides was observed a significant effect (p <0.05) independent of diluent and treatment, for ejaculated spermatozoa with minor defects and normal. From the potassium concentration was showed that only the vigor suffered group effect (p <0.05). In the spermatozoa from caudal epididymis, fructose was a significant effect for group interaction, treatment and time on motility, for the vigor there was a significant interaction effect between treatment and time and group effect, in the fructose and potassium concentrations, respectively. For the chloride concentration was observed a significant interaction between group and treatment on the occurrence of major defects, (p <0.05). After 24 hours of storage, the best values were observed in sperm without seminal plasma from caudal epididymis, however in the presence of seminal plasma, sperm from the ejaculate withstood better conservation (p <0.05). The addition of seminal plasma reduced seminal parameters of the sperm from caudal epididymis. The biochemical composition of seminal plasma of goats raised in northeastern Brazil interferes directly on the quality of cooled semen. |

Os ejaculados de 8 animais foram coletados pela manhà e o sÃmen fresco foi dividido de forma a se obter 4 tratamentos: CG (lavado e nÃo lavado), TG (lavado e nÃo lavado). No mesmo dia à tarde, os animais foram castrados para obtenÃÃo dos espermatozoides da cauda do epidÃdimo, dos quais foram retiradas quatro alÃquotas de 100 L. Em duas delas foi adicionada 120 L de plasma seminal autÃlogo, constituindo assim outros 4 tratamentos: CG (espermatozoide da cauda do epidÃdimo com e sem plasma seminal) e TG (espermatozoide da cauda do epidÃdimo com e sem plasma seminal). O plasma seminal foi avaliado quanto à composiÃÃo bioquÃmica. As amostras dos 8 tratamentos foram analisadas quanto ao vigor, motilidade e morfologia espermÃtica, a 0 h (fresco) e apÃs 2 e 24 h conservaÃÃo a 5ÂC. Os dados foram analisados pelo SAS@. No agrupamento pela concentraÃÃo de frutose e cloretos foi observado um efeito significativo (p<0,05) independente do diluidor e tratamento, para os espermatozoides ejaculados com defeitos menores e normais. A partir da concentraÃÃo de potÃssio foi revelado que apenas o vigor sofreu efeito de grupo (p<0,05). Nos espermatozoides da cauda do epidÃdimo, a frutose constituiu efeito significativo para interaÃÃo entre grupo, tratamento e tempo sobre a motilidade, jà para o vigor foi observado um efeito significativo da interaÃÃo entre tratamento e tempo e efeito de grupo, para as concentraÃÃes de frutose e potÃssio respectivamente. Para a concentraÃÃo de cloretos, foi constatado um efeito significativo na interaÃÃo entre o grupo e o tratamento sobre a ocorrÃncia dos defeitos maiores, (p<0,05). ApÃs a conservaÃÃo por 24 horas, os melhores valores foram observados nos espermatozoides da cauda do epidÃdimo sem plasma seminal, no entanto na presenÃa de plasma seminal, os espermatozoides do ejaculado suportaram melhor a conservaÃÃo. A adiÃÃo de plasma seminal reduziu os parÃmetros seminais dos espermatozoides da cauda do epidÃdimo.
