Extraction, characterization and toxicity of pectin noni (Morinda citrifolia L)

Resumo: Noni (Morinda citrifolia L.) is a plant that has aroused the interest of the scientific community. However there are few scientific papers about the pectin extracted from this fruit. The extraction was conducted at three pHâs range (3, 7 and 10), within a time and temperature interval. Three different extracting agents were used. The pectin extracted at pH 3 (PDN-a) was subjected to the process of sulfation and confirmed by elemental analysis. All pectin samples from extraction processes and citrus pectin of commercial use (VetecÂ) were characterized by FT - IR, where it was possible to determine the degree of esterification (30%) which was confirmed with the results obtained by potentiometric titration and 1H NMR. Through the GPC and viscosity analyzes it was possible to determine the respective molecular weights of the pectin samples (3.8 x105 6.8 x105 g / mol). The NMR 1H analysis were important in the determination of the five signals corresponding to hydrogens of the galacturonic acid chain. The thermal analysis, TGA and DSC, observed the range of stability of pectins and compared them in relation to citrus pectin of commercial use. In AFM analysis it was possible to observe the molecular level of how the structure of pectin was affected (fragmented) by extraction and sulfation process. In the Zeta potential analyzes it was confirmed the anionic character of the pectin chain (-32.2 mV and -42.1 mV) to the pectin extracted at acid pH and their sulfated sample. It was determined Surface Tension and Critical Micelle Concentration (CMC) for the pectin sample (pH 3). The rheological study and some physicochemical properties such as moisture content, determination of lipids, determination of protein and determination of soluble fiber were also performed. The PDN-a sample which obtained the highest yield (19%) and its polymer chain was little affected by the extraction process, was subjected to in vivo toxicological tests (mice), which was carried an acute oral toxicity test, determination of hematological and biochemical parameters and check of the wet relative weight of organs, where its toxicity has been dropped. PDN-s (sulfated pectin) was used for in vitro cytotoxicity testing (human blood) and verification of the enzyme lactate dehydrogenase activity, where there was no significant differences compared to controls used.

Todas as amostras de pectinas provenientes dos processos de extraÃÃes e a pectina cÃtrica de uso comercial (VetecÂ) foram caracterizadas por: FT â IR, onde foi possÃvel determinar o grau de esterificaÃÃo (30%) o qual foi confirmado com os resultados obtidos por titulaÃÃo potenciomÃtrica e por RMN 1H. AtravÃs das anÃlises de GPC e de viscosidade foi possÃvel determinar respectivamente as massas molares das amostras de pectinas (3,8 x105 a 6,8 x105 g/mol). As anÃlises de RMN de 1H foram importantes para determinaÃÃo dos cinco sinais correspondentes aos hidrogÃnios da cadeia do acido galacturÃnico. Jà com as anÃlises tÃrmicas, TGA e DSC, foi possÃvel observar a faixa de estabilidade das pectinas e comparar em relaÃÃo à pectina cÃtrica de uso comercial. Nas anÃlises de AFM foi possÃvel observar a nÃvel molecular como a estrutura da pectina foi afetada (fragmentada) pelo processo de extraÃÃo e sulfataÃÃo. Nas anÃlises do Potencial Zeta foi confirmado o carÃter aniÃnico da cadeia de pectina (-32,2 mV e -42,1 mV), para a pectina extraÃda em pH Ãcido e sua amostra sulfatada. Foi determinada a TensÃo Superficial e a ConcentraÃÃo Micelar CrÃtica (CMC), para a amostra de pectina (pH 3). O Estudo reolÃgico e algumas propriedades fÃsico-quÃmicas, como, Teor de umidade, DeterminaÃÃo de lipÃdios, DeterminaÃÃo de proteÃnas e DeterminaÃÃo de fibras solÃveis tambÃm foram realizadas. A amostra PDN-a que obteve um melhor rendimento (19%) e sua cadeia polimÃrica foi pouco afetada pelo processo de extraÃÃo, foi submetida a testes toxicolÃgicos in vivo (ratos), onde foi realizado um ensaio de toxicidade aguda via oral, DeterminaÃÃo dos parÃmetros hematolÃgicos e bioquÃmicos e verificaÃÃo do peso Ãmido relativo dos ÃrgÃos, onde sua toxicidade foi descartada.
