Resumo: | Mammary neoplasm is the third most common cancer in feline, with most of the time a malignant behavior. For a diagnostic evaluation, prognosis and therefore the direction of a medical-surgical conduct appropriate classification is an essential characteristic. But often this classification is difficult, and used other techniques as an aid. Specialized techniques such as immunohistochemistry, to evaluate cell proliferation and apoptosis can serve as a basis for the diagnosis and prognosis. The application of these parameters helps comparative studies of feline mammary carcinogenesis and human. Thus, this study evaluated the hypothesis that the feline malignant mammary tumors have high expression of Ki-67 and cleaved caspase 3. The main objective was to evaluate the anatomic-pathological features and expression of Ki-67 and cleaved caspase-3 in malignant breast neoplasms domestic cats by linking the results of histopathology and immunohistochemistry. A total of 30 cases of mammary tumors in cats. The material was fixed in buffered formalin 10% and subsequently processed and embedded in paraffin. The tumors were classified according to World Health Organization, through histopathological examination of samples stained with hematoxylin and eosin. The reaction of immunohistochemistry was performed with the primary antibodies anti-Ki-67 and anticaspase 3. Most animals were classified as mixed breed cats, with a mean age of 10.43 years. Microscopically, we observed the presence of hyperchromatic nucleus, evident nucleoli, moderate pleomorphism and mitotic figures. In the cases studied, the neoplastic cells showed pattern-forming tubular, papillary, cystic and / or solid. Of the 30 samples were observed one case of tubulo-papillary adenocarcinoma with squamous differentiation, two cases of cystic tubular adenocarcinomas, five cases of tubular adenocarcinomas, papillary cystic, five cases of solid tubular adenocarcinomas, eight cases of tubular adenocarcinomas, papillary and nine cases of simple tubular adenocarcinomas. In three cases (3/13) showed regional lymph node metastasis. In nine cases (9/24), the surgical margins were invaded by neoplastic cells in two cases (2/24) there was invasion of muscle tissue and in one case (1/24) there was invasion to adipose tissue. In feline malignant mammary tumors was studied immunoreactivity of anti-Ki-67 and anti-cleaved Caspase 3, but there was no positive correlation between them. Although few studies with feline mammary tumors, mainly correlating the proliferative activity and apoptotic tumor. Para uma avaliação diagnóstica, prognóstica e consequentemente no direcionamento de uma conduta clínico-cirúrgica adequada, a classificação é uma ferramenta essencial. Porém, muitas vezes essa classificação se torna difícil, sendo utilizadas outras técnicas como um auxílio. Técnicas especializadas como a imuno-histoquímica, para avaliação da proliferação celular e apoptose podem servir como base para o diagnóstico e prognóstico. A aplicação desses parâmetros auxilia estudos comparativos da carcinogênese mamária felina e humana. Desta forma, nesse estudo foi avaliada a hipótese de que as neoplasias mamárias malignas felinas apresentam elevada expressão de Ki-67 e de caspase 3 clivada. O objetivo principal foi avaliar os aspectos anatomo-histopatológicos e a expressão imuno-histoquímica de Ki-67 e de caspase-3 clivada em neoplasias mamárias malignas de felinos domésticos, associando os resultados do exame histopatológico e imunohistoquímico. Foram utilizadas 30 amostras de tumores mamários em gatas. O material foi fixado em formol tamponado a 10% e posteriormente processado e incluído em parafina. As neoplasias foram classificadas de acordo com a Organização Mundial de Saúde, por meio de exame histopatológico de amostras coradas em Hematoxilina e Eosina. A reação de imuno-histoquímica foi realizada com os anticorpos primários anti-Ki-67 e anti-caspase 3. A maioria dos animais foram classificados como felinos sem raça definida, apresentando média de faixa etária de 10,4 anos. Microscopicamente, observou-se a presença de núcleo hipercromático, nucléolos evidentes, pleomorfismo moderado e figuras de mitose. Nos casos estudados, as células neoplásicas apresentaram-se formando padrão tubular, papilar, cístico e/ou sólido. Das 30 amostras analisadas, obteve-se um diagnóstico de adenocarcinoma túbulo-papilífero com diferenciação escamosa, dois de adenocarcinomas tubulares císticos, cinco de adenocarcinomas túbulospapilíferos císticos, cinco de adenocarcinomas tubulares sólidos, oito de adenocarcinomas túbulos-papilíferos e nove de adenocarcinomas tubulares simples. Em nove amostras (9/24), as margens cirúrgicas encontravam-se invadidas por células neoplásicas, em duas (2/24) observou-se invasão de tecido muscular e em uma amostra (1/24) houve invasão para tecido adiposo. Observou-se metástase em linfonodo regional em três amostras estudadas (3/13). Nas neoplasias mamárias malignas felinas estudadas houve imunoreatividade dos anticorpos anti-Ki-67 e anti-Caspase 3 clivada, porém não foi observada correlação positiva entre ambos. Ainda são escassos os trabalhos com neoplasia mamária felina, principalmente, correlacionando a atividade proliferativa e apoptótica tumoral. Através deste trabalho, foi possível analisar o comportamento histopatológico e imuno-histoquímico das neoplasias mamárias felinas malignas.
