MicroRNA como biomarcador na insuficiência cardíaca

Resumo: Introduction: Heart Failure (HF) is characterized as the inability to maintain the metabolic needs of tissues when the heart fails to maintain adequate cardiac output. In the cardiovascular system approximately 150 to 200 microRNAs are expressed, regulated in response to cardiac stress and part of it can be found in the bloodstream being termed as circulating miRNAs. Objective: To evaluate the profiles of circulating miRNAs in patients with chronic heart failure with reduced ejection fraction. Methods: Cross-sectional study with 10 patients: 05 patients with HF with EF <50% and 05 of the control group. 10 ml of blood was collected for miRNAs analysis, centrifuged at 4 ° C for 10 min and then stored at -80 ° C until isolation. The MiRNeasy Serum / Plasma Kit (Qiagen, Hilden, Germany) containing the QIAzol reagent for the extraction of total RNA and MiRNeasy Serum / Plasma Spike-In Control for standardization of serum microRNA purification was used to purify the total RNA. Expression of the microRNAs was determined by the microarray technique using the GeneChip ™ miRNA 4.0 Affymetrix® template and the analysis and quantification were performed by Affymetrix® GeneChip® Command Console® (Affymetrix miRNA 4.0). Results: A mean ejection fraction in patients with HF was 39.6%. The expression of miRNAs was increased for mir-1207-5P (P = 0.005), mir-6887 (p = 0.007), mir-548a-3p (p = 0.03), mir-4530 P = 0.02), and reduced expression for mir-3157-3p (p = 0.02) and mir-1234-3p (0.04). Conclusion: In this study, new circulating miRNAs were identified in patients with heart failure with reduced ejection fraction, to be confirmed in new studies.

No sistema cardiovascular são expressos aproximadamente 150 a 200 microRNAs, regulados em resposta ao estresse cardíaco e parte dele podem ser encontrados na corrente sanguínea sendo denominados como miRNAs circulantes. Objetivo: Avaliar os perfis de miRNAs circulantes em pacientes com insuficiência cardíaca crônica com fração de ejeção reduzida. Metodologia: Estudo transversal com 10 pacientes: 05 portadores de IC com FE < 50% e 05 do grupo controle. Foram coletados 10 ml de sangue para análise dos miRNAs, centrifugado à 4Cº por 10 min e depois armazenado a -80ºC até o momento do isolamento. Para purificação do RNA total foi utlizado o Kit MiRNeasy Serum / Plasma (Qiagen, Hilden, Alemanha), contendo o reagente QIAzol para extração do RNA total e MiRNeasy Serum / Plasma Spike-In Control para normalização da purificação de microRNA do soro. A expressão dos microRNAs foi determinado pela técnica de microarranjo utilizando a matriz GeneChip™ miRNA 4.0 Affymetrix® e a análise e quantificação foram realizadas pelo Software Affymetrix® GeneChip® Command Console® (Affymetrix miARN 4.0). Resultados: A média de fração de ejeção nos paciente com IC foi de 39,6%. Na análise dos miRNAs obteve-se expressão aumentada para o mir-1207-5P (P=0,005), mir-6887 (p= 0,007), mir-548a-3p (p=0,03), mir-4530 (p= 0,02), e expressão reduzida para o mir-3157-3p (p= 0,02) e mir-1234-3p (0,04).
