São conhecidos como agentes ultramicroscópicos, parasitas intracelulares obrigatórios e dependentes da maquinaria celular hospedeira para sua multiplicação. Entretanto, nas últimas décadas, ganhou projeção científica um grupo monofilético de vírus, extremamente diversificado e complexo, com uma extraordinária variedade estrutural e genômica, levantando questionamentos a respeito da origem e natureza dos vírus. Este grupo é conhecido como Vírus Gigantes Núcleo-Citoplasmáticos de DNA (NCLDV), ou mais recentemente a ordem proposta Megavirales. Nos anos 2000, foi caracterizado um vírus extremamente complexo que fez questionar-se tudo o que se conhecia a respeito dos vírus. Este vírus foi denominado Acanthamoeba polyphaga mimivirus, o qual foi inserido em uma família viral própria, Mimiviridae, por não se encaixar em nenhuma família já existente. Recentemente, vários vírus gigantes têm sido isolados a partir de diferentes ambientes e espécimes, desta forma, o objetivo deste trabalho foi avaliar a presença de isolados de mimivírus em amostras de ostras e água do mar coletadas em três regiões litorâneas brasileiras. Neste estudo foram utilizadas técnicas de enriquecimento das amostras e isolamento em monocamadas celulares de espécies de Acanthamoeba castellanii. Para a caracterização biológica dos novos isolados de mimivirírus foram utilizados ensaios de microscopia eletrônica de transmissão, de resistência quando expostos à luz ultravioleta e a temperatura, além do uso de técnicas moleculares a fim de avaliar o perfil molecular de cada isolado. Quatro novos isolados foram caracterizados biológica e molecularmente, nos quais foi possível observar partículas virais semelhantes aos vírus pertencentes à família Mimiviridae, bem como possíveis diferenças biológicas e/ou estruturais após a exposição aos ensaios de resistência. Os novos isolados pertencem ao grupo A da família Mimiviridae. Além disso, nossos resultados sugerem que as ostras são uma excelente fonte para o isolamento de vírus gigantes.Viruses are ubiquitous organisms found in any environment in which life is present. In the 2000s, the characterization of an extremely complex virus put in question everything that was known about viruses. This virus, named Acanthamoeba polyphaga mimivirus, was placed into its viral own family, Mimiviridae, not fitting to any existing family. Furthermore, several additional giant viruses have been isolated from different environments and specimens. In this context, the aim of this study was to evaluate the incidence of isolated mimivirus in samples of oysters and sea water collected in three brazilian coastal regions. We used techniques of samples enrichment for subsequent isolation in cell monolayers species of Acanthamoeba castellanii. For biological characterization of new isolates we used electron microscopy transmission and assays of stress when exposed to ultraviolet light and temperature, and we used molecular techniques to evaluate the molecular profile of each isolate. Four new isolates have been characterized biological and molecularly in which it was possible to observe viral particles similar to viruses belonging to the family Mimiviridae and possible biological and / or structural differences after exposure to the stress tests. The new isolates belong to the group A Mimiviridae family.
