As sequências encontradas através do sequenciamento de nova geração RNA-seq e montagem do transcriptoma pelo programa Trinity, foram submetidas à anotação gênica para que fossem identificadas por meio de buscas de similaridade contra diferentes bancos de dados. Em seguida, foram classificadas em categorias funcionais e avaliadas quanto a sua função biológica a partir de análise filogenética. A identificação correlacionada aos compostos da peçonha favorece melhor compreensão dos processos fisiopatológicos do envenenamento, bem como ferramentas para melhorias nos métodos de profilaxia e tratamento para o Loxoscelismo. Nesta dissertação objetivamos nossos estudos transcriptômicos nas proteínas da peçonha de L. laeta responsáveis pela principal atividade tóxica do composto, que são as Fosfolipases D, esfingomielinases D ou proteínas dermonecróticas. Nesse trabalho, foram descritas 19 sequências de fosfolipases D potencialmente encontradas na peçonha de L. laeta. A partir da análise e descrição da expressão das fosfolipases D da peçonha e interatividade molecular, torna-se possível o aperfeiçoamento de ferramentas para o estudo dos mecanismos de ação da atividade dermonecrótica dessas enzimas. Além disso, o estudo aprofundado da expressão dessas enzimas na peçonha e sua constituição favorecem futuros estudos sobre a filogenia de suas proteínas.This study aims to analyze, by bioinformatic tools, the transcripts of the brown spider venom gland Loxosceles laeta to expand the phospholipase D database. Sequences found through the new generation of sequencing RNA-seq and transcriptome assembly of the program Trinity, were subjected to gene annotation that so they could be identified through searches of similarity against different databases. Then they were classified into functional categories and had their biological function evaluated from a phylogeneticstandpoint. The identification correlated to the venom of compounds promotes better understanding of the pathophysiological processes of envenomation cases, as well as tools for improvements in methods of prophylaxis and treatment for loxoscelism. This dissertation aimed to describe the venom proteins responsible for its major toxic activities, which are the PhospholipaseD, Sphingomyelinase D or Dermonecrotic Proteins. In this study, 19 putative Phospholipase D sequences were described from the venom gland of the spider L. laeta. From the analysis and description of the Phospholipases D expression from the venom and their molecular interaction, it is possible to improve tools for the study of mechanisms of action of dermonecrotic activity of these enzymes.
