A ifecção é um processo dependente de diversos fatores como as glicoproteínas presentes na superfície do parasito e da célula-alvo, a população de T.cruzi e a sinalização celular desencadeada pela interação parasito-célula-alvo. Sabe-se que, em modelos experimentais, o perfil anatomopatológico e imunológico da infecção depende da população de T. cruzi. Neste trabalho propusemo-nos a analisar se a citocina pró-inflamatória TNF-alfa exerce efeito sobre a infecção de células epiteliais por tripomastigotas de clones de T. cruzi, pertencentes a DTU (Discret Typing Units) distintas. Também investigamos se as células infectadas mostram alguma alteração ultraestrutural no período de 22 horas de infecção. Nossos resultados mostram que células epiteliais LLC-MK2 e HEK293 são mais susceptíveis à infecção pelo clone Col1.7G2 pertencente a DTU/TCI, quando comparadas com a infecção pelo clone CL-Brener pertencente a TCVI. Em relação ao clone CL-Brener, as células HEK293 são mais susceptíveis à infecção que as células LLC-MK2. Nossos dados mostram também que, ao contrário do descrito anteriormente para a cepa Y, o TNF-alfa não exerce efeito sobre a proporção de células infectadas e sobre o número de amastigotas intracelulares nos tipos celulares estudados. De modo interessante, a análise em microscopia eletrônica de transmissão indica que as mitocôndrias das células epiteliais tendem a se acumular ao redor do parasito, muitas vezes se justapondo à sua membrana plasmática, no período de 22 horas de infecção. Estes dados demonstram a complexidade do processo de invasão, que se mostrou dependente do tipo celular e da população do parasito utilizada. Desse modo, torna-se importante a continuidade de estudos para detecção de moléculas envolvidas neste processo, bem como o envolvimento de mitocôndrias no processo de infecção e permanência do parasito na célula hospedeiraTrypanosoma cruzi, the etiologic agent of Chagas disease, is able to enter into all cell types of mammalian cells. It is known that, in experimental models, the pathological and immunological profile of the infection depends on the population of T. cruzi. In this work, we aimed to examine whether the pro-inflammatory cytokine TNF-alpha has an effect on epithelial cell invasion by trypomastigotes of two populations of T. cruzi belonging to different DTU (Discrete Typing Units). We have also investigated whether the infected cells show any ultrastructural changes after 22 hours of infection. Our results show that epithelial LLC-MK2 cells and HEK293 cells are more susceptible to infection by Col1.7G2 clone, which belongs to the DTU/TCI class compared with infection by clone CL-Brener, which belongs to TCVI. In relation to clone CL-Brener, HEK293 cells are more susceptible to infection than the LLC-MK2 cells. Our data also show that, contrary to the previously findings with strain Y, TNF-alpha had no effect on the proportions of infected cells and the number of intracellular parasites in both cell types studied. Interestingly, analysis by transmission electron microscopy indicates that mitochondria of epithelial cells tend to accumulate around the parasite, often in close juxtaposition to its plasma membrane, during the period of 22 hours of infection. These data demonstrate the complexity of the invasion process, which proved to be dependent on the cell type and the population of the parasite.
