Efeito imuno-metabólico do extrato aquoso da Roupala montana (Proteácea) em modelo experimental

Resumo: Diabetes Mellitus is characterized by a chronic metabolic disorder characterized by hyperglycemia and disorder in the metabolism of lipids, carbohydrates and proteins, resulting from the non-production, little production and misuse of the insulin, and with vascular consequences, which are closely intertwined. This has become a growing epidemic in the last century, with the exponential increase in obesity, poor eating habits and other factors. For many years people use medicinal herbs to control blood glucose, by popular culture or information without any proof, or just an alternative to the synthetic drugs available today.That being said, this work aimed to assess the effect of the aqueous extract of the Roupala montana species tree (Proteaceae), in male Winstar lineage rats, on the biochemical and immunological profiles and on some toxic response. The animals were randomly divided into four groups (n total 24), control group – rats with standard diet; sucrose group – standard diet and water with 10% sucrose; sucrose and treated group - 10% sucrose and extract gavage; and treated group – only extract gavage, all fed with standardized ration. The extract was administered via gavage at a concentration of 250mg/kg throughout the study daily for five consecutive weeks. All subjects had access to water and food ad libitum. Parameters such as body weight, water intake, feed consumption and blood glucose were evaluated weekly. After five weeks, the animals were anesthetized and sacrificed to obtain blood and weigh the organs. With the blood, analyzes of biochemical parameters and quantification of oxidized lipoprotein via monoclonal antibody IgG assay were performed. No major changes were observed in the groups tested, only a slight increase in the ALT enzyme in those using sucrose, and a high in triglycerides in those using sucrose. There was no change in the weight of the organs of the animals. There was water consumption in the animals and feed consumption was reduced in the groups using sucrose and a slight decrease in the group treated with extract. There were no changes in the experimental lipoprotein profiles.

There were no changes in the experimental lipoprotein profiles.O Diabetes Mellitus se caracteriza pela desordem metabólica crônica caracterizada pela hiperglicemia e desordem no metabolismo de lipídeos, carboidratos e proteínas, decorrentes da não produção, pouca produção e má utilização do hormônio insulina e com consequências vasculares, que estão intimamente interrelacionadas. Este tornou-se uma epidemia crescente no ultimo século, com o aumento exponencial da obesidade, maus hábitos alimentares e outros fatores. Por muitos anos povos fazem a utilização de plantas medicinais para o controle da glicemia, por cultura popular ou informações sem alguma comprovação, ou apenas uma alternativa aos medicamentos sintéticos hoje disponíveis. Diante disso, este trabalho visou avaliar o efeito do extrato aquoso da arvore da espécie Roupala montana (Proteaceae), em ratos machos da linhagem Winstar, nos perfis bioquímicos, imunológicos e alguma resposta tóxica. Os animais foram divididos aleatoriamente em quatro grupos (n total 24), grupo controle – ratos com dieta padrão; grupo sacarose – dieta padrão e água com sacarose a 10%; grupo sacarose e tratado - sacarose a 10% e gavage do extrato; e grupo tratado – somente gavage do extrato, todos alimentados com ração padronizada. A administração do extrato foi feita via gavage na concentração de 250mg/kg durante todo o estudo diariamente por cinco semanas consecutivas. Todos animais tiveram acesso a água e ração ad libitum. Foram avaliados parâmetros como peso corpóreo, ingestão hídrica, consumo de ração e glicemia semanalmente. Após as cinco semanas os animais foram anestesiados e sacrificados para obtenção do sangue e pesagem dos órgãos. Com o sangue foram feitas analises de parâmetros bioquímicos e quantificação de lipoproteína oxidada e Apob-D via ensaio de anticorpo monoclonal IgG. Não foram observadas grandes mudanças nos parâmetros testados, apenas uma leve alta na enzima ALT nos grupos em uso de sacarose, e alta nos triglicérides no grupo em uso de sacarose. Não houve alterações no peso dos órgãos dos animais. Houve alteração no consumo hídrico nos animais em uso de sacarose, e consumo de ração foi diminuído nos grupos em uso de sacarose, e leve decréscimo no grupo tratado com extrato.
