Resumo: Depression and anxiety are psychiatric disorders and present psychological, behavioral and physiological symptoms. As for anxiety and depression treatment, the major problem found is the deficiency on drugs that associate the high effectiveness, high bioavailability and low side effects. Therefore, the L-ascorbil-6-palmitato (PA) associated with nanoparticles could be an inovative alternative for these disorders treatment. In this context, the objective of this study was to evaluate the antidepressant- and anxiolytic-like effects of AP-loaded nanocapsules in different depression and anxiety models on mice. Moreover, we verified the involvement of Na+, K+ -ATPase activity in brain. Animals were divided in 3 groups; group I: control; group II: nano and group III: free. Animals were pretreated intragastrically (i.g) by 24, 48, 72 or 96 hours before of behavioral tests. Group I were pretreated with suspensions containing blank nanocapsules, group II received AP-loaded nanocapsules (30 mg/kg) and group III received free PA (30 mg/kg). In order to evaluate animals locomotor and exploratory activity it was taken the open field test. The forced swimming and tail suspension tests ware used in order to evaluate the antidepressive-type effect, while the elevated plus maze and light/dark box tests were used in order to verify the anxiolytic-type effect. After behavioral tests, animals were killed and brains were removed to determinate Na+, K+ -ATPase activity. Results showed that pretreatment of 24 from 96 hours with AP-loaded nanocapsules reduced the immobility time in forced swim, while free PA in reduced the immobility time with 24 and 48 hours of pretreatment. In the tail suspension tests, AP-loaded nanocapsules and free form reduced the immobility time of 24 to 96 hours of pretreatment. In the elevated plus maze, there was an increase of the number of jump in the pretreated of 48 hours with AP-loaded nanocapsules. No change was observed in the other parameters and groups in this test. In the light/dark box test, all time of pretreatment with AP-loaded nanocapsules increased the latency time for the first entry in the dark compartment and time that the animal remained in the light compartment. Furthermore, no change was found in the Na+, K+-ATPase activity in brains of mice. In the open field test there was no change in animals locomotor and exploratory activity in any of the times tested and groups analyzed. Therefore, we concluded that the AP-loaded nanocapsules showed anxiolytic-like effect. Moreover, AP-loaded nanocapsules and free form presented antidepressant-like effect, but the effect of nanocapsules was more prolonged. Na+, K+-ATPase activity was not involved in the pharmacological effects.

Quanto ao tratamento da ansiedade e da depressão, o maior problema encontrado é a deficiência de fármacos que associem alta eficácia, alta biodisponibilidade e baixos efeitos colaterais. Assim, o L-ascorbil-6-palmitato (PA) associado às nanopartículas poderia ser uma alternativa inovadora para o tratamento desses transtornos. Neste contexto, o presente estudo teve como objetivo avaliar o efeito tipoantidepressivo e ansiolítico de nanocápsulas contendo PA, em diferentes modelos de depressão e ansiedade em camundongos. Além disso, foi verificado o envolvimento da atividade da enzima Na+, K+-ATPase cerebral no efeito farmacológico. Os animais foram divididos em 3 grupos: grupo I: controle; grupo II: nano e grupo III: livre. Os pré-tratamentos dos animais foram 24, 48, 72 ou 96 h antes da realização dos testes comportamentais, pela via intragástrica (i.g). Os animais do grupo I foram pré-tratados com suspensões contendo nanocápsulas brancas, do grupo II receberam PA nanoencapsulado (30 mg/kg) e do grupo III foram pré-tratados com o PA na forma livre (30 mg/kg). Para avaliar a atividade locomotora e exploratória dos animais foi realizado a teste do campo aberto. Os testes do nado forçado e da suspensão da cauda foram utilizados para avaliar o efeito tipo-antidepressivo, enquanto os testes do labirinto em cruz elevada e do claro/escuro foram utilizados para verificar o efeito tipo-ansiolítico. No final dos testes comportamentais, os animais foram sacrificados e os cérebros removidos para verificar a atividade da enzima Na+, K+-ATPase. Os resultados desse estudo demonstraram uma redução do tempo de imobilidade no teste do nado forçado nos animais pré-tratados com nanocápsulas de PA nos tempos de 24 a 96 h, enquanto o PA na forma livre reduziu o tempo de imobilidade apenas nos pré-tratamentos de 24 e 48 h. Enquanto que no teste da suspensão da cauda, tanto o fármaco nanoencapsulado quanto na forma livre apresentaram uma redução no tempo de imobilidade de 24 a 96 h. No teste da cruz elevada, houve um aumento no número de espiadas nos animais pré-tratados 48 h com as nanocápsulas de PA. Os outros parâmetros e os demais grupos analisados neste teste não apresentaram alterações. No teste do claro-escuro, as nanocápsulas de PA aumentaram o tempo de latência para a primeira entrada do animal no compartimento escuro em todos os tempos de pré-tratamentos e também aumentaram o tempo que o animal permaneceu no compartimento claro. No teste do campo aberto não houve modificação na atividade locomotora e exploratória dos animais, em nenhum dos tempos testados e grupos analisados.Além disso, nenhuma diferença significativa foi encontrada na atividade Na+, K+-ATPase em nenhum dos tempos de pré-tratamento no cérebro dos camundongos. Com isso, pode-se concluir que as nanocápsulas contendo PA apresentaram efeito tipo-ansiolítico, enquanto o PA na forma livre não apresentou esse efeito. Além disso, tanto o PA na forma livre quanto o encapsulado apresentaram efeito tipo-antidepressivo, porém o efeito das nanocápsulas de PA foi mais prolongado.
