Resumo: Non-steroidal anti-inflammatory drugs (NSAIDs) are used in the treatment of pain, fever and inflammatory processes. However, these drugs have some adverse effects on the gastrointestinal tract. Indomethacin (INDO) may cause adverse reactions to the patient, such as gastric erosions and ulcers. Oxidative stress occurs when the cellular antioxidant defense mechanism is no longer able to inactivate reactive oxygen species (ROS) or excess production. Oxidative stress leads to severe damage to DNA, which has been associated with several pathologies, including gastric ulcers. Antioxidants are able to fight these reactive species, thus protecting biomolecules and reducing disease risk. Flavonoids are the plant metabolites that provide protection against ROS. Naringin (NA) and naringenin (NG) are flavonoids responsible for the bitter taste of citrus fruits. They have low solubility in water, they undergo first pass metabolism and low bioavailability when administered orally. Therefore, the importance of nanoencapsulate these flavonoids, to increase stability, changing the pharmacokinetics and biodistribution of these.Nanocapsule suspensions were prepared by the interfacial deposition technique of the preformed polymer. Eight male Wistar rats, divided in control group, omeprazole, NA + NG (L), white nanocapsule (NB), NG + NA (NN) nanocapsule were used. For the induction of ulcer, 48 mg INDO / kg body mass were administered by orogastric gavage. For the pre-treatment groups, omeprazole (5mg / kg body weight) and L (0.5mg of each active, NA and NG), NB (0.5mg of each active, NA and NG) were administered via Oral / day for 28 days. On the 28th day, INDO was administered by v.o. One hour after treatments. All rats were fasted 24 hours prior to induction of gastric injury by INDO. Four hours after INDO administration, rats were euthanized and their stomachs dissected and separated for histological and biochemical analyzes to calculate the gastric and preventive ulcer index, pH determination and oxidative stress parameters, to verify if there was protection By the action of nanocapsules. Pretreatment with NN nanocapsules proved to be more effective than omeprazole in preventing INDO-induced ulcer. More studies are needed so that these assets can be used in therapy.

Porém, estes fármacos como, por exemplo, a indometacina (INDO), podem apresentar alguns efeitos adversos no trato gastrointestinal como erosões gástricas e úlceras. As úlceras gástricas vêm sendo associadas ao desequilíbrio redox na mucosa gástrica, ou seja, quando a ação de espécies reativas se sobrepõe à ação dos mecanismos de defesa antioxidante. Este desequilíbrio redox é denominado estresse oxidativo. Com o objetivo de restaurar o equilíbrio redox, e reduzir o risco do aparecimento de úlceras na mucosa gástrica, temse proposto o uso de substâncias antioxidantes como a naringina (NA) e a naringenina (NG) presentes em frutas cítricas. Porém, tanto NA quanto NG possuem baixa solubilidade em água, sofrem metabolismo de primeira passagem e têm baixa biodisponibilidade quando administradas por via oral. Para contornar estes problemas, nanoencapsular estes flavonoides, pode aumentar a estabilidade, e biodistribuição dos mesmos. Nesse contexto, foram preparadas as suspensões de nanocápsulas com NA e NG, através da técnica de deposição interfacial do polímero pré-formado. Para o desenvolvimento do presente estudo, foram utilizados 80 ratos Wistar machos divididos randomicamente em grupos com e sem úlcera. Os grupos sem úlcera foram controle (C), omeprazol (O), NA + NG livres (L), nanocápsula branca (NB), nanocápsula com NA + NG (NN). Os grupos com úlcera foram controle (U), omeprazole (UO), NA + NG livre (UL), nanocápsula trançada (UNB) e nanocápsula com NA + NG (UNN). Para a indução de úlcera, 48 mg de INDO/kg de massa corporal foram administrados por sonda gástrica orogástrica. Todos os grupos foram tratados oralmente durante 28 dias com os pré-tratamentos. As dosagens foram 5 mg/kg de massa corporal de omeprazol e 0,5 mg/massa corporal de cada uma, NA e NG nos grupos L e NN. No 28º dia, para os grupos com úlcera, a INDO foi administrada por v.o., 1 h após a administração dos tratamentos. Todos os ratos estavam em jejum de 24 horas antes da indução de lesão gástrica por INDO. Quatro horas após a administração de INDO, os ratos foram eutanaziados e os seus estômagos dissecados e separados para as análises histológicas, bioquímicas, para cálculo do índice de úlcera gástrica e preventivo, a determinação do pH e parâmetros do estresse oxidativo O pré-tratamento com as nanocápsulas NN se mostrou mais eficaz que o omeprazol em relação à prevenção da úlcera induzida por INDO, indicando que houve certa prevenção pelas nanopartículas.
