Resumo: The carqueja-gaucha is an endemic species of Rio Grande do Sul, poorly explored and needs further research in micropropagation. In recent years, increased interest in using the technique of micropropagation of medicinal and aromatic plants, which provides a large-scale production. The objective was to evaluate the effect of different concentrations of sucrose, salts of MS medium, nitrogen and different growth regulators on in vitro cultivation of gorse. The explants were nodal segments with two axillary buds from plants already established in vitro. In the 1st experiment were tested with different concentrations of sucrose in MS medium (1% 3% 5% and 7%). In the 2nd experiment tested whether variations in concentrations of salts (macro and micronutrients) of the MS (MS / 3, MS / 2, MS; 2MS). In the 3rd experiment tested changes in the concentrations of nitrogen (N, N / 2 N / 3 N / 4) in MS medium. In the 4th experiment evaluated different concentrations (BAP): M1 (MS), M2 (MS +0 +0.05 GA3 BAP), M3 (MS +0.10 +0.05 GA3 BAP), M4 (MS + BAP +0.30 GA3 0.05 mg L-1). The pH was adjusted to 5.8 before adding agar at a concentration of 8 g L-1 and subsequently autoclaved at 121 º C for 20 minutes. The experimental design was completely randomized design with four replicates of five explants per flask. We evaluated the numbers of leaves, shoots and roots and callus formed at 15 and 30 days of cultivation. In a second experiment, one can observe a greater number of leaves in a medium containing 5% sucrose and within 30 days. But the basic MS medium with normal concentration of sucrose 3% showed a lower number of leaves at 30 days of cultivation. The highest number of shoots was observed in medium with 1% sucrose for 30 days. With increasing concentration decrease in the production of shoots. In the 2nd experiment, it was found that the number of leaves was stimulated by increasing the salt concentration of MS medium, media containing 100% and 200% of the MS culture medium, can also be observed an increase in root presence over time and not for the variations of salts in MS medium. In the 3rd experiment leaf number was greater in medium containing the lowest concentration of nitrogen and over time there was an increase in their number, the number of shoots was lower in the usual concentration of nitrogen and 30 days was an increase of shoots. In the 4th experiment all doses tested BAP promoted an increase in shoots of carqueja-gaucha and 30 days showed a higher number of shoots. In the medium with growth regulators had an increase in the production of callus at the base of the explant with increasing time in culture.

Nos últimos anos, aumentou o interesse da utilização da técnica de micropropagação em plantas aromáticas e medicinais, o que proporciona uma produção em larga escala. O objetivo do trabalho foi avaliar o efeito de diferentes concentrações de: sacarose, sais do meio MS, nitrogênio e diferentes reguladores de crescimento no cultivo in vitro de carqueja. Como explantes foram utilizados segmentos nodais com duas gemas axilares, oriundos de plantas já estabelecidas in vitro. No 1º experimento foram testadas as diferentes concentrações de sacarose no meio MS (1%, 3% 5% e 7%). No 2º experimento testou-se as variações das concentrações de sais (macro e micronutrientes) do meio MS (MS/3; MS/2; MS; 2MS). No 3º experimento foram testadas variações nas concentrações de nitrogênio (N, N/2, N/3, N/4) no meio MS. No 4ª experimento foram avaliadas diferentes concentrações de BAP (Benzilaminopurina): M1 (MS); M2 (MS+0BAP+0,05GA3);M3(MS+0,10BAP+0,05GA3); M4 (MS+0,30BAP+0,05GA3 mg L-1). O pH foi ajustado para 5,8 antes da adição do ágar na concentração de 8 g L-1 e posteriormente autoclavado a 121 ºC por 20 minutos. O delineamento experimental foi inteiramente casualizado, com quatro repetições de cinco explantes por frasco. Foram avaliados os números de folhas, brotos e calos e raízes formadas aos 15 e 30 dias de cultivo. No 1º experimento, pode-se observar um maior número de folhas no meio contendo 5% de sacarose, no período de 30 dias. Já o meio MS básico com concentração usual de sacarose 3% apresentou um menor número de folhas aos 30 dias de cultivo. O maior número de brotos foi observado no meio com 1% de sacarose aos 30 dias. Com o aumento da concentração houve decréscimo na produção de brotos. No 2º experimento, verificou-se que o número de folhas foi estimulado pelo aumento da concentração de sais do meio MS, nos meios contendo 100% e 200% dos sais do meio MS; também pode-se observar um aumento da presença de raízes com o passar do tempo e não em relação às variações de sais no meio MS. No 3º experimento, o número de folhas foi maior no meio contendo a menor concentração de nitrogênio e, com o passar do tempo, houve um aumento de número das mesmas, o número de brotações foi menor na concentração usual de nitrogênio e aos 30 dias observou-se aumento de brotações. No 4º experimento, todas as doses de BAP testadas promoveram um aumento de brotações de carqueja-gaúcha e aos 30 dias apresentou um maior número de brotações.
