Defesa: Curitiba, 24/03/2010Bibliografia: fls. 70-97Área de concentraçao: Exercício e esporteResumo: Diabetes Mellitus (DM) é um grupo de disfunções metabólicas resultantes de defeitos na secreção da insulina, na ação da insulina ou de ambas. Indivíduos diabéticos apresentam maior suscetibilidade a infecções e a doenças cardiovasculares. O treinamento intervalado envolve períodos curtos há longos de exercícios repetidos com alta intensidade, intercalado com períodos de recuperação. Assim, nosso estudo objetivou avaliar o impacto do treinamento intervalado sobre parâmetros bioquímicos sangüíneos e função de células imunitárias em ratos diabéticos do tipo I. Diabetes experimental foi induzida em ratos da linhagem Wistar machos adultos através da injeção intravenosa de estreptozotocina (60 mg/kg). Após a constatação do quadro diabético (7 dias após a inoculação) os animais foram divididos em 4 grupos: sedentário (SE), treinamento intervalado (TI), diabético (DM), diabético treinamento intervalado (DTI). O treinamento intervalado de natação consistiu de 30 segundos de exercício com 30 segundos de recuperação, 15% da massa corporal do animal, freqüência de 4 vezes por semana, duração de 30 minutos, durante 6 semanas. Foram analisados tempo total de exercício, massa corporal total, glicemia, triacilglicerolemia, colesterolemia e parâmetros da funcionalidade de macrófagos obtidos da cavidade peritoneal dos ratos e neutrófilos sanguíneos, como a fagocitose de zimosan, retenção de vermelho neutro, produção de ânion superóxido (O2 -) e de peróxido de hidrogênio (H2O2) (EROS). Proliferação de linfócitos mesentéricos estimulados com Concanavalina A também foi avaliada. O tempo total do treinamento intervalado foi 60% e 25% menor do que as recomendações da ADA e da ACSM. Diabetes experimental resultou em perda de peso, hiperglicemia, colesterolemia e triacilglicerolemia. O treinamento intervalado reverteu estas alterações aumentando a massa corporal total em 19,2% do grupo DTI, reduziu a massa corporal do grupo TI em 14,88%, reduziu a hiperglicemia (21%), colesterolemia (41%) e triacilglicerolemia (43%) do grupo DTI. Diabetes experimental aumentou 94% afagocitose de neutrófilos, como também, EROs (72% e 33%, para ânion e superóxido) e 1,4 vezes o volume lisossomal, enquanto que o treinamento intervalado não provocou alteração adicional nestes parâmetros. Já na funcionalida e de macrófagos o diabetes experimental aumentou a fagocitose em 2,7 vezes, volume lisossomal em 92%, EROs (2 vezes e 7,5%, aniôn e superóxido), adesão 30%. Com o treinamento intervalado o grupo DTI obteve um aumento adicional (1,6 vezes) na fagocitose e o grupo TI aumentou 3,8 vezes, além disto, houve aumento no volume lisossomal (91%), EROs (2 vezes e 8%, para ânion e superóxido) e 18% para adesão no grupo TI. Com relação a proliferação de linfócitos, houve aumento basal de 74% dos linfócitos obtidos dos animais não diabéticos treinados (TI). A estimulação com Con A aumentou a proliferação dos linfócitos T dos animais diabéticos e não diabéticos, induzindo a um aumento adicional de 46% no grupo treinado (DTI). Nossos resultados sugerem que o treinamento intervalado melhora o perfil lipídico e glicemia reduzindo o risco de doenças cardiovasculares e melhora a funcionalidade das células imunitárias de diabéticos, promovendo maior proteção contra infecções.Abstract: Diabetes Mellitus (DM) is a group of dysfunctions metabolic resultants of defects in the secretion of the insulin, in the action of the insulin or of both. Diabetic subjects presents larger susceptibility to nfections and cardiovascular diseases. The interval training involves short periods there is long of repeated exercises with high intensity, inserted with recovery periods. Thereby, our study aimed to evaluate the impact of the interval training on blood biochemical parameters and immune cells function in diabetic rats of the type I. Experimental diabetes was induced in male Wistar rats through streptozotocin (60mg/kg) intravenous injection. After diabetic (7 days after induction), status animals were divided into 4 groups: sedentary (SE), diabect (DI), interval training (IT) and diabetic interval training (DIT). The training swimming interval consisted of 30 seconds f exercise with 30 seconds of recovery, 15% of the body weight of the animal, frequency of 4 times a week, duration of 30 minutes, for 6 weeks. It was measured total ime of exercise, total body mass, glycemia, triacylglycerolemia, colesterolemia and some parameters of immune cells function like zimosan phagocytosis, lysosomal retention, superoxide and hydrogen peroxide production (ROS). Macrophage were obtained from peritoneal cavity and neutrophils from the blood. Mesenteric lymphocyte proliferation stimulated with Concanavalin A were also evaluated. The total time of the interval training was 60% and 25% smaller than the recommendations of ADA and ACSM. Experimental diabetes resulted in weight loss, hiperglycemia, colesterolemia and hypertriacylglycerolemia. The interval training reverted these alterations increasing the total body mass in 19,2% of the group DTI, it reduced the corporal weight of the group IT (14,88%), it reduced the hiperglycemia (21%), colesterolemia (41%) and triacylglycerolemia (43%) of the group DTI. Experimental diabetes increased neutrophils phagocytosis (94%), as well as, ROS (72% and 33%, for anion and superoxide) and 1,4 times the lysosomal retention, while the interval training didn't provoke additional alteration in these parameters. Macrophage functionality the experimental diabetes increased the phagocytosis (2,7 times), lysosomal volume 92%, ROS (2 times and 7,5%, anion and superoxide), adhesion 30%. With the interval training the group DTI obtained an additional increase (1,6 times) in the phagocytosis and the group IT increased 3,8 times, besides, there was increase in the lysosomal volume (91%), ROS (2 times and 8%, for anion and superoxide) and 18% for adhesion in the group TI. Regarding proliferation of lymphocytes, there was basal increase of 74% of the obtained ymphocytes of the animals no trained diabetics (IT). The stimulation with Con A increased lymphocytes proliferation of the diabetic animals and no diabetics, inducing to an additional increase of 46% in the trained group (DTI).
