Agentes da theileriose e da anaplasmose granulocítica em equídeos de microrregiões do estado do Rio de Janeiro

Resumo: | Este trabalho foi realizado nas microrregiões de Itaguaí e Serrana no estado do Rio de Janeiro para estudar os agentes da theileriose e da anaplasmose granulocítica equina. Amostras de soro e sangue foram submetidas à reação de imunofluorescência indireta (RIFI), reação em cadeia da polimerase em tempo real (RT-PCR) e a análise hematológica. A prevalência geral para T. equi foi de 81,09% (n=579), com maior prevalência (p<0,05) para microrregião de Itaguaí (85,43%), quando comparado a Serrana (76,92%). A região, altitude, nível da propriedade e origem dos equinos foram associados (p<0,05) com a soropositividade para T. equi. Equinos criados na microrregião de Itaguaí apresentaram 11% mais chance (RP=1,11; p=0,003) de terem contato com T. equi comparado aqueles criados na microrregião Serrana. Além disso, quando são criados a altitudes abaixo de 500m apresentaram 10% de chance a mais abaixo de 500m (RP=1.10; p=0,014) de se tornarem soropositivos. Equinos criados em propriedades de nível ruim (RP=1,13; p=0,018) e nascidos na propriedade (RP=1,10; p=0,008) apresentaram mais chance de terem contato com T. equi. A frequência de equinos soropositivos para A. phagocytophilum foi de 17% (n=17) das amostras examinadas no município de Seropédica. Apenas a idade apresentou associação (p=0,044) com a soropositividade dos equinos para A. phagocytophilum. Das 350 amostras avaliadas pela RT-PCR, 0,86% (n=3) apresentaram amplificação específica para o gene msp2. As amostras positivas apresentaram valores médios do CT de 35,09±1,48 ciclos e de Tm de 78,56±0,53ºC, semelhante a do controle positivo, de 78,45±0,32ºC. As microrregiões de Itaguaí e Serrana são endêmicas para theileriose equina e de estabilidade enzoótica para T. equi. Apenas os fatores relacionados à área de coleta influenciam a soropositividade dos equinos para T. equi nas microrregiões estudadas. A presença de anticorpos anti-A. phagocytophilum e a evidência molecular da infecção por esse agente em equinos do município de Seropédica, no estado do Rio de Janeiro, sugere a circulação de A. phagocytophilum nessa região. |

Blood and serum samples were subjected to indirect fluorescent antibody test (IFAT), real time polymerase chain reaction (RT-PCR) and hematological analysis. The overall prevalence for T. equi was 81.09% (n=579), with higher prevalence (p<0.05) for Itaguaí microregion (85.43%), when compared to Serrana (76.92%). The region, altitude, level of farm and origin of horses were associated (p<0.05) with seropositivity for T. equi. Horses reared in the Itaguaí microregion had 11% more likely (PR=1.11, p=0.003) of having contact with T. equi compared to those reared in the Serrana microregion. Moreover, when are raired at altitudes below 500m showed 10% chance less (PR=1.10; p=0,014) of becoming seropositive. Horses reared in poor level farms (PR=1.13, p=0.018) and born on the farm (PR=1.10, p=0.008) showed more chance of having contact with T. equi. The main ticks found attached in horses were Amblyomma cajennense and Dermacentor (Anocentor) nitens. The frequency of seropositive horses for A. phagocytophilum was 17% (n=17) of the samples tested in the Seropédica county. No association (p>0.05) was observed among the studied factors and seropositivity of horses to A. phagocytophilum. Of 350 samples tested by RT-PCR, 0.86% (n=3) showed specific amplification for msp2 gene. The positive samples showed average CT value of 35.09±1.48 cycles and Tm of 78.56±0.53°C, similar to the positive control, 78.45±0.32°C. The Itaguaí and Serrana microregions are endemic for equine theileriosis and of enzootic stability for T. equi. Only factors related to collection area influenced the seropositivity of horses for T. equi in the microregions studied. Este trabalho foi realizado nas microrregiões de Itaguaí e Serrana no estado do Rio de Janeiro para estudar os agentes da theileriose e da anaplasmose granulocítica equina. Amostras de soro e sangue foram submetidas à reação de imunofluorescência indireta (RIFI), reação em cadeia da polimerase em tempo real (RT-PCR) e a análise hematológica. A prevalência geral para T. equi foi de 81,09% (n=579), com maior prevalência (p<0,05) para microrregião de Itaguaí (85,43%), quando comparado a Serrana (76,92%). A região, altitude, nível da propriedade e origem dos equinos foram associados (p<0,05) com a soropositividade para T. equi. Equinos criados na microrregião de Itaguaí apresentaram 11% mais chance (RP=1,11; p=0,003) de terem contato com T. equi comparado aqueles criados na microrregião Serrana. Além disso, quando são criados a altitudes abaixo de 500m apresentaram 10% de chance a mais abaixo de 500m (RP=1.10; p=0,014) de se tornarem soropositivos. Equinos criados em propriedades de nível ruim (RP=1,13; p=0,018) e nascidos na propriedade (RP=1,10; p=0,008) apresentaram mais chance de terem contato com T. equi. A frequência de equinos soropositivos para A. phagocytophilum foi de 17% (n=17) das amostras examinadas no município de Seropédica. Apenas a idade apresentou associação (p=0,044) com a soropositividade dos equinos para A. phagocytophilum. Das 350 amostras avaliadas pela RT-PCR, 0,86% (n=3) apresentaram amplificação específica para o gene msp2. As amostras positivas apresentaram valores médios do CT de 35,09±1,48 ciclos e de Tm de 78,56±0,53ºC, semelhante a do controle positivo, de 78,45±0,32ºC. As microrregiões de Itaguaí e Serrana são endêmicas para theileriose equina e de estabilidade enzoótica para T. equi. Apenas os fatores relacionados à área de coleta influenciam a soropositividade dos equinos para T. equi nas microrregiões estudadas. A presença de anticorpos anti-A. phagocytophilum e a evidência molecular da infecção por esse agente em equinos do município de Seropédica, no estado do Rio de Janeiro, sugere a circulação de A. phagocytophilum nessa região. |
