Estudo fitoqu?mico do caule de Talinum triangulare (Portulacaceae) e propriedades antioxidante, quelante e de inibi??o da enzima tirosinase

Resumo: The chemical screening of the chloroformic and methanolic fractions revealed phenols, flavonoids, cathequines and the presence of other components, indicating the special metabolites of the chiquimic acid and acetate metabolic role. The T. triangulare total phenols was determined by colorimetric method according to Folin-Ciocalteu pattern indicating, after statistic treatment, a larger phenols concentration in the hydromethanolic fraction when compared to the crude extract. UV spectroscopy was used for the evaluation of Fe II complexes formation with polar extract of T. triangulare stem. The results were compared to the commercial sample of Ginkgo biloba, indicating a similar pattern to the clinically used phytotherapic. The crude extract and hydromethanolic fraction antioxidant assays, performed using the DPPH method. The tyrosinase inhibitory effect was also evaluated indicating the hydromethanolic fraction as more effective, in same way as the antioxidant effect. The phytochemical study of the chloroformic fraction permitted the isolation and characterization of the methyl octadecanoate (1), and the new (3E)-4-methyl-3- {[(octanoyloxy)acetyl]oxy}methyl)-3-heptenoic acid (2) and olean-9,12-dien-3-(?-Dglucopyranosyl)- 28,29-dimethyl ester (3), by spectroscopic methods such as NMR 1H and 13C, IR and GC-MS.

Al?m disso, foram avaliados a atividade antioxidante e o efeito inibit?rio frente a enzima tirosinase do extrato bruto e da fra??o hidrometan?lica. Finalmente foi realizada a an?lise fitoqu?mica da fra??o clorof?rmica. A prospec??o qu?mica feita com as fra??es clorof?rmica e hidrometan?lica revelou a presen?a de fen?is, flavon?ides, catequinas e outros constituintes que caracterizaram metab?litos da via do ?cido chiqu?mico e do acetato. A quantidade de fen?is totais em T. triangulare foi determinada colorimetricamente, conforme o procedimento padr?o de Folin-Ciocalteu e atrav?s de tratamento estat?stico foi poss?vel determinar a sua concentra??o no extrato bruto e na fra??o hidrometan?lica, sendo observado maior concentra??o nesta ?ltima. A espectofotometria no UV-vis?vel foi usada para avaliar a possibilidade de complexa??o do extrato polar do caule de T. triangulare com ?ons de Fe II. Os resultados obtidos foram comparados com os da amostra comercial de Ginkgo biloba, onde se observou que a esp?cie T. triangulare apresentou efeito semelhante ao do fitoter?pico Ginkgo biloba usado clinicamente. Os testes da atividade antioxidante com o extrato bruto e a fra??o hidrometan?lica, realizado atrav?s do m?todo DPPH, e da a??o inibit?ria frente a enzima tirosinase, indicaram de forma comparativa, que em ambos a fra??o hidrometan?lica apresentou maior potencial que o extrato bruto.
