Resumo: | The treatment of pain mainly of orofacial pain and its therapeutic interventions in order to improve the quality of life and remedy the painful symptoms of the patient have been the main current problem of medicine, not only by limiting the options that There are, but also by the amount of side effects that accompany them. Therefore, the study of substances derived from natural products is a hope for the improvement of pain management. The species of plants of the genus Agave or "sisal" used in the popular medicine of the Brazilian Northeast for possessing favorable therapeutic properties, have already been used by the industry as a basis for synthesis of steroidal hormones. Previous studies have observed that hecogenin, a steroidal saponin present in this genus of plants, in the acetylated form, demonstrated anti-nociceptive and anti-inflammatory effect in animal models. In order to overcome the low aqueous solubility of HA, it has been envisaged to take advantage of the ability of cyclodextrin (CD) molecules to form inclusion complexes with non-polar substances in order to improve water solubility and hence bioavailability. Objective: To evaluate the anti-nociceptive activity of the Hecogenina acetate (HA) and the AH/β-cyclodextrin (HA-ΒCD) in orofacial pain model in rodents. Methods: Swiss male mice (25-30 g) (CEPA/UFS # 04/12) subjected to orofacial pain models induced by formalin, capsaicin and glutamate. The animals (n = 6) were treated with the HA or HA-ΒCD (20 mg/kg), or vehicle, orally. The results were considered significant when p < 0.05. Results: It was observed that the HA-ΒCD complex presented an improvement in the anti-nociceptive effect in the formalin-induced and glutamate pain model compared to the control (p < 0.01 and p < 0.001). Conclusion: With the obtained results, therefore, we observed that the HA when complexed with a β-CD showed anti-nociceptive effect in animal models of orofacial use. |

Por isso o estudo de substâncias derivadas de produtos naturais são uma esperança para a melhoria do manejo da dor. As espécies de plantas do gênero Agave ou “sisal” utilizadas na medicina popular do nordeste brasileiro por possuírem propriedades terapêuticas favoráveis, já vem sendo utilizadas pela indústria como base para síntese de hormônios esteroidais. Estudos anteriores observaram que a hecogenina, uma saponina esteroidal presente nesse gênero de plantas, na forma acetilada, demonstrou efeito anti-nociceptivo e anti-inflamatório em modelos animais. A fim de contornar a baixa solubilidade aquosa do AH, pensou-se em aproveitar a capacidade que as moléculas de ciclodextrina (CD) possuem em formar complexos de inclusão com substâncias de caráter apolar, a fim de melhorar a hidrossolubilidade e consequentemente a biodisponibilidade. Objetivo: Avaliar a atividade anti-nociceptiva do acetato de hecogenina (AH) e do AH/β-ciclodextrina (AH-βCD) em modelo de dor orofacial em roedores. Métodos: Foram utilizados camundongos Swiss machos (25-30 g) (CEPA/UFS#04/12) submetidos a modelos de dor orofacial induzida por formalina, capsaicina e glutamato. Os animais (n=6) foram tratados com o AH ou AH-βCD (20 mg/kg), ou veículo, por via oral. Os resultados foram considerados significativos quando p< 0,05. Resultados: Foi observado que o complexo AH-βCD apresentou melhoria no efeito anti-nociceptivo no modelo de dor induzido por formalina e glutamato em comparação com o controle (p<0,01 e p<0,001).
