Resumo: | The complexity of compounds present in plants allows them to be used for different diseases, which is due to the presence of bioactive compounds named as secondary metabolites. Considering the lack of chemical knowledge of Croton grewioides species and the need to establish new methodologies for the extraction of its antioxidante compounds, in this work was carried out the optimization of the extraction processes by ultrasound and microwave using an experimental planning, aided by statistical analysis. Thus, it was possible to determine the best extraction conditions for each of the technique studied, ultrasound-assisted extraction (UAE) and microwave assisted extraction (MAE). From the best conditions of extraction, the extracts of C. grewioides were analyzed by liquid chromatography and their fingerprint chromatograms submitted to chemometric analysis. High resolution mass spectrometry analyzes were also carried out, using the Orbitrap analyzer, with the objective of obtaining a chemical profile of mass spectrums, chemical profile of the extracts, and try to chemically identify the molecular formula and propose the chemical structure to the largest number possible of compounds present in this species. Through the chemometric results from both fingerprint, the identified metabolites and the antioxidant activities obtained, it was possible to assigned that the best extraction technique for antioxidant compounds from C. grewioides is microwave assisted extraction. Additionally, it was possible to propose, for the first time, structures of forty one compounds in the extracts of this species from the analysis of high resolution mass spectrometry, being considered an unprecedented method for the chemical study of this species. |

Considerando a escassez no conhecimento químico da espécie Croton grewioides e a necessidade de se estabelecer novas metodologias de extração de seus compostos antioxidantes, neste trabalho foi realizado a otimização dos processos de extração por ultrassom e micro-ondas utilizando um planejamento experimental, auxiliados por tratamentos estatísticos. Foi possível a determinação das melhores condições de extração para cada uma das técnicas estudadas, a extração assistida por ultrassom (EAU) e a extração assistida por micro-ondas (EAM). Determinadas as melhores condições de extração, os extratos de C. grewioides foram analisados por cromatografia líquida e seus cromatogramas fingerprint submetidos a análise quimiométrica. Também foram feitas análises por espectrometria de massas de alta resolução, utilizando o analisador Orbitrap, com o objetivo de se obter os espectros de massas perfil químico dos extratos, atribuir a fórmula molecular e propor a estrutura química ao maior número possível de compostos presentes nesta espécie. Através dos resultados quimiométricos dos dois fingerprints, dos metabólitos identificados e dos dados de atividades antioxidantes obtidos, foi possível atribuir que a melhor técnica de extração para compostos antioxidantes a partir de C. grewioides é a extração assistida por micro-ondas.
