Germinação e estabelecimento inicial in vitro de Lavandula angustifolia

Resumo: | The plant tissue culture techniques includes a set in which each explant (cell, tissue or organ) is isolated under aseptic conditions on artificial nutrient medium. This process is based on the principle of totipotentiality cells, or any plant organism cell has all the genetic information necessary for the regeneration of a whole plant. Nevertheless, some plant species have difficulty germination is the case of Lavandula angustifolia, and an unevenness in the development of seedlings and plants in the field, due to natural variability resulting from sexual propagation. In this context, the in vitro culture can solve or minimize hot spots in seedling establishment with medicinal properties and production of essential oils or by micropropagation. Thus, three experiments were carried out in order to study the germination and early development of this species. In the first experiment evaluated the efficiency of use of hydrogen peroxide in the pretreatment of seed treated with different concentrations of gibberellic acid in vitro culture: 0.0; 1.0; 2.0 and 3.5 mg L-1. Then, we assessed the influence of using conventional lamps and growlux MS and LS media at concentrations of 50 and 100% of the salts. Finally, an assay which evaluated the effect of the new class of regulators comprising polyamines putrescine, spermine and spermidine concentrations in different under conventional lamps. In all experiments biometric characteristics such as number of leaves were evaluated, shoot length and root fresh weight, percentage of abnormal seedlings, survival rate and the kind of germination parameters such as germination, emergence rate, start time, end and average germination, sync, uncertainty. Best results were found when retracted the seeds with hydrogen peroxide and inoculated in medium containing 2.5 mg L -1 of gibberellic acid. The use of growlux lamps did not differ across conventional light bulbs in the evaluated characteristics, but it was identified that the species grows best in LS means . As for the polyamines found a generally favoring the germination, shoot and leaf number of lavender seedlings at 0.5 mg L-1 spermine. except when applied spermine generating fewer sheet in this condition. |

Este processo baseia-se no princípio da totipotencialidade das células, ou seja, qualquer célula de organismo vegetal apresenta todas as informações genéticas necessárias à regeneração de uma planta completa. Algumas espécies vegetais entretanto apresentam dificuldade de germinação, como é o caso da Lavandula angustifolia, além de uma desuniformidade no desenvolvimento das mudas e das plantas à campo, devido à variabilidade natural resultante da propagação sexuada. Neste contexto, o cultivo in vitro pode resolver ou minimizar pontos críticos comuns no estabelecimento de plantas com propriedades medicinais e ou produtoras de óleos essenciais pelo processo de micropropagação. Dessa forma, foram realizados três experimentos no intuito de estudar o estabelecimento in vitro e desenvolvimento inicial dessa espécie. No primeiro experimento avaliou-se a eficiência da utilização de peróxido de hidrogênio no pré-tratamento de sementes inoculadas em meio de cultura com diferentes concentrações de ácido giberélico em cultivo in vitro: 0,0; 1,0; 2,0 e 3,5 mg L-1. No segundo, avaliou-se a influencia do uso das lâmpadas growlux e convencional em meios MS e LS nas concentrações de 50 e 100% dos sais. E por fim, avaliou-se o efeito da classe de reguladores de crescimento poliaminas que compreendem a putrescina, espermina e espermidina em diferentes concentrações. Em todos os experimentos foram avaliadas características biométricas como número de folhas, comprimentos de parte aérea e radicular, massa fresca, porcentagem de plântulas anormais, e os parâmetros da germinação da espécie como porcentagem de germinação e velocidade de emergência. Melhores resultados foram encontrados quando se aplicou pre- tratamento com peróxido de hidrogênio nas sementes com a posterior inoculação em meio contendo 2,5 mg L-1 de ácido giberélico. O uso de lâmpadas growlux não apresentou diferença frente as lâmpadas convencionais nas características avaliadas, porém foi identificado que a espécie desenvolve melhor em meio LS.
