Esta atividade ? importante para o meio ambiente, pois realizam a degrada??o da mat?ria org?nica depositada sobre o solo em plantios florestais ou em florestas naturais. Para produ??o de a??cares ferment?veis ou bioenergia, ? utilizado biomassa celul?sica, sendo considerada uma alternativa ben?fica, ao qual necessita da produ??o enzim?tica de micro-organismos. O presente trabalho tem como objetivo, identificar e analisar a atividade enzim?tica de fungos de um banco de micro-organismos celulol?ticos oriundos de solo e serapilheira sob plantio de eucalipto. A identifica??o foi realizada para cinco fungos, por meio de t?cnicas biomoleculares, como: extra??o de DNA, quantifica??o visual em gel de agarose, PCR e sequenciamento de DNA. Foram selecionados dois fungos celulol?ticos, que atenderam os crit?rios para a an?lise de atividade enzim?tica de beta-glicosidase e endoglucanase, submetidos a meio l?quido de cultivo, in vitro, de CMC (carboximetilcelulose) e de p? de madeira de eucalipto. Os cinco fungos identificados s?o ascomicetos, onde 4 apresentaram homologia acima de 99%, sendo pertencentes aos g?neros: Penicillium para IM1-12, Simplicillium para IM32-12, Exophiala para IM7-21, Rhinocladiella para IM1-58, e g?nero n?o definido para IM1-34B, evidenciando a diversidade do banco gen?tico estudado. A produ??o de beta-glicosidase e endoglucanase alcan?aram, respectivamente, valores de 0,109 U.mL-1 e 0,587 U.mL-1 para o isolado IM32-12, e 0,090 U.mL-1 e 0,505 U.mL-1 para o isolado IM1-12. O meio de cultivo com CMC foi mais eficiente para a produ??o enzim?tica. O isolado IM32-12 apresentou maior valor para produ??o enzim?tica tanto de atividade de beta-glicosidase quanto de endoglucanase, nos diferentes meios de cultivo, demonstrando maior potencial enzim?tico em rela??o ao isolado IM1-12. Contudo, recomenda-se novos estudos voltados para a otimiza??o da produ??o enzim?tica desses micro-organismos, com intuito de promover o m?ximo potencial enzim?tico.Submitted by Jos? This activity is important for the environment, since they degrade the organic matter deposited on the soil in forest plantations or in natural forests. For the production of fermentable sugars or bioenergy, cellulosic biomass is used, being considered a beneficial alternative, and qualified by the enzymatic production of microorganisms. The present work aims to identify and analyze the enzymatic activity of fungi from a bank of cellulolytic microorganisms from soil and litter under eucalyptus plantation. The identification was carried out for five fungi, using biomolecular techniques, such as: DNA extraction, visual quantification in agarose gel, PCR and DNA sequencing. Two cellulolytic fungi were selected, which met the criteria for the analysis of enzymatic activity of beta-glycosidase and endoglucanase, submitted to liquid culture medium, in vitro, of CMC (carboxymethyl cellulose) and eucalyptus wood powder. The five fungi identified are ascomycetes, where 4 showed homology above 99%, belonging to the genera: Penicillium for IM1-12, Simplicillium for IM32-12, Exophiala for IM7-21, Rhinocladiella for IM1-58, and gender not defined for IM1-34B, showing the diversity of the genetic bank studied. The production of beta-glucosidase and endoglucanase reached values of 0.109 U.mL-1 and 0.587 U.mL-1, respectively, for IM32-12 alone, and 0.090 U.mL-1 and 0.505 U.mL-1 for the IM1-12 isolated. The CMC culture medium was more efficient for enzymatic production. The isolated IM32-12 showed higher value for enzymatic production of both beta-glucosidase and endoglucanase activity, in the different culture media, showing greater enzymatic potential in relation to the isolated IM1-12.
