No entanto, para garantir a viabilidade econ?mica do processo de convers?o de biomassa em combust?vel, ? fundamental reduzir o custo da etapa de hidr?lise da biomassa. O uso de micro-organismos para a hidr?lise e para a convers?o de celulose em etanol tem chamado a aten??o de empresas que atuam no desenvolvimento de enzimas, em especial o complexo celulase. Diante disso, o presente trabalho teve como objetivo selecionar micro-organismos potenciais produtores de celulases. Para isso, amostras de solos e serapilheira sob floresta de eucalipto em diferentes idades de plantio foram coletadas e submetidas ao isolamento de micro-organismos em meio de cultura seletivo. Escolheu-se solos em ?reas de cultivo de eucalipto devido o objetivo ser selecionar micro-organismos que atuem, especificamente, em madeira de eucalipto. Foram isolados 870 micro-organismos para an?lises com o m?todo de descolora??o de vermelho congo. 336 fungos filamentosos, 8 bact?rias e 3 leveduras foram selecionados com capacidade de sintetizar enzimas celulol?ticas. Destes, 163 foram avaliados como bons produtores de celulases, devido apresentarem ?ndices enzim?ticos superior a 2,5. Destaca-se que os isolados 25?50, 1?74 e 1?75 apresentaram elevados ?ndices enzim?ticos: 24,0; 22,0 e 20,0, respectivamente. A partir desses dados, esses isolados podem ser estudados molecularmente e o gene envolvido na produ??o dessa enzima ser caracterizado e clonado. Os resultados obtidos no presente trabalho mostram a import?ncia da prospec??o de novas linhagens de micro-organismos produtores de celulases. De maneira geral, pode-se concluir que ? poss?vel a utiliza??o de t?cnicas de screening para selecionar linhagens de um extenso banco de isolados. However, to ensure the economic viability of the biomass conversion process into fuel, it is critical to reduce the cost of the biomass hydrolysis step. The use of microorganisms for the hydrolysis and conversion of cellulose into ethanol has attracted the attention of companies involved in the development of enzymes, especially the cellulase complex. Therefore, the present work aimed to select potential micro-organisms producing cellulases. For this, samples of soils and litter under eucalyptus forest at different planting ages were collected and submitted to the isolation of microorganisms in selective culture medium. Soils were selected in areas of eucalyptus cultivation due to the objective being to select microorganisms that specifically act on eucalyptus wood. 870 microorganisms were isolated for analysis with the congo red discoloration method. 336 filamentous fungi, 8 bacteria and 3 yeasts were selected with the ability to synthesize cellulolytic enzymes. Of these, 163 were evaluated as good producers of cellulases, due to their enzymatic indexes higher than 2.5. It is noteworthy that isolates 25-50, 1-74 and 1-75 showed high enzymatic indices: 24.0; 22.0 and 20.0, respectively. From these data, these isolates can be molecularly studied and the gene involved in the production of that enzyme is characterized and cloned. The results obtained in the present work show the importance of the prospection of new lines of microorganisms producing cellulases. In general, it can be concluded that it is possible to use screening techniques to select strains from an extensive database of isolates.
