Ultra-estrutura e transcriptoma de urócitos de pupas de Melipona quadrifasciata (Hymenoptera: Meliponini)

Resumo: | The fat body of adult bees consists of different cell types as trophocytes and oenocytes, however, in young pupae some trophocytes are modified in cells called urocytes whose function remains unknown in bees. This study evaluated the morphology, transcriptome and the viability to maintain primary culture of urocytes in the stingless bee Melipona quadrifasciata. The urocytes of M. quadrifasciata are spherical cells with an irregular surface and white color when observed under the stereomicroscope. May present individually or in groups associated with tracheae. The urocytes had a single core, small, spherical and centered. This work revealed the cytosol of urocytes neutral polysaccharides, lipid droplets, vesicles protein, calcium and urate granules containing vesicles. This suggests that they function as excretory cells in bee pupae. The cells of the fat body urate bee M. quadrifasciata were purified and their RNA were extracted for cDNA library construction. We performed PCR of cDNA fragments and these were fractionated on a column for separation of fragments larger than 200 bp. This was followed by the protocol binding fragments > 200bp to the vector pTZ57R / T and subsequent cloning. Plasmid DNA was extracted from clones and sent to automated sequencing. The sequences obtained were analyzed using the BLASTn and BLASTx of the NCBI. Most sequences obtained did not find homology with the data available for bees, as well as to other organisms, resulting in no significant findings for the development of knowledge about the transcripts obtained for urocytes in this work. |

Este trabalho avaliou a morfologia de urócitos na abelha Melipona quadrifasciata, bem como a possibilidade de manutenção dessas células em cultivo primário. Os urócitos de M. quadrifasciata são células esféricas de superfície irregular e coloração branca quando observadas ao estereomicroscópio. Podem se apresentar individualizados ou em grupos associados a traqueias. Os urócitos apresentaram um único núcleo, pequeno, esférico e centralizado. O presente trabalho revelou no citosol dos urócitos polissacarídeos neutros, gotas lipídicas, vesículas de proteínas, cálcio e vesículas contendo grânulos de urato. Isto sugere que eles funcionem como células excretoras em pupas da abelha. As células de urato do corpo gorduroso da abelha M. quadrifasciata foram purificadas e tiveram seu RNA extraído para construção da biblioteca de cDNA. Foi realizada a PCR dos fragmentos de cDNA e esses foram fracionados em coluna para separação de fragmentos maiores que 200 pb. Seguiu-se o protocolo de ligação dos fragmentos >200pb ao vetor pTZ57R/T e posterior clonagem. O DNA plasmidial foi extraído dos clones e enviado para sequenciamento automático. As sequências obtidas foram analisadas utilizando-se o BLASTn e BLASTx do NCBI.
