Resumo: Cardiovascular diseases are the main cause of death in the Western world. They have as the main morphological change the formation of atheromatose plates. Epidemiological and experimental studies demonstrate beneficial action from anti-oxidants, such as the grape polyphenols and the [alfa]-tocopherol, on the development of these diseases. With these speculations this work was developed with the objective of verifying the effecf of the food supplement with grape polyphenols and the [alfa]-tocopherol on the atherosclerosis evolution in Apo E-/- mice. Five-week old mice received an atherogenic diet for six weeks. After this time they were divided into four groups: Control received an atherogenic diet and water, Grape , received an atherogenic diet and grape extract; Tocopherol , received an atherogenic diet supplemented with 400 UI/ Kg of a diet of [alfa]-tocoferol acetate and water ; and the Grape and Tocopherol , which received an atherogenic diet supplemented with 400 UI/ Kg of a diet of [alfa]-tocopherol acetate and grape extract. The treatment lasted for eleven weeks. On the final week the feces were collected. After sacrificing the mice (euthanasia) we extracted the blood, aorta, heart and liver. We measured total cholesterol, triacilgliceroids, total polyiphenols from the serum, liver and feces. In the serum and liver we measured vitamin E and hydroperoxide lipids. In the liver and feces we analysed the fatty acids profile. The heart and aorta were processed histologically and a morphological- morphometric analysis was done. The treatments showed considerable hypocholesterolemic effect with decrease of the seric and hepathic levels of cholesterol. There was an increase of fecal excretion of cholesterol with the supplementation with anti-oxidants. There was no change in the triacilgliceroids, polyphenols and malondialdeid on the hepathic tissue of the animals. The formation of hydroperoxide lipids showed a trend of being less when the animals received both anti-oxidants. The supplemented groups showed greater levels of vitamin E seric and hepathic. On the fatty acids profile we did not observe any statistical difference, however it is noticeable a greater trend in preserving polyinsaturated fatty acids when there was a supplementation of grape polyphenols. The morphologic analysis revealed a lower level of evolution of the animals atheromatose plates that received [alfa]-tocopherol. There was no difference in the size of the larger damage found. The results suggest that the supplementation with both anti-oxidants obtained a better effect on the evolution of the atherosclerosis since it lowered the levels of cholesterol, important risk factor, and it improved the anti-oxidant potential on the animals organism.

Têm como principal alteração morfológica a formação de placas ateromatosas. Estudos epidemiológicos e experimentais demonstram ação benéfica de antioxidantes, como os polifenóis da uva e o [alfa]-tocoferol, sobre o desenvolvimento dessas doenças. Diante dessas especulações o presente trabalho foi desenvolvido com o objetivo de verificar o efeito da suplementação alimentar com polifenóis da uva e [alfa]-tocoferol sobre a evolução da aterosclerose em camundongos Apo E -/-. Camundongos com cinco semanas de vida receberam dieta aterogênica durante seis semanas. Após esse período foram divididos em quatro grupos: Controle , recebeu dieta aterogênica e água; Uva , recebeu dieta aterogênica e extrato de uva; Tocoferol , recebeu dieta aterogênica suplementada com 400 UI/ Kg de dieta de acetato de α-tocoferol e água; e Uva e Tocoferol , que recebeu dieta aterogênica suplementada com 400 UI/ Kg de dieta de acetato de α-tocoferol e extrato de uva. O tratamento prosseguiu por onze semanas. Na última semana as fezes foram recolhidas. Após eutanásia foram retirados sangue, aorta, coração e fígado. Foram dosados colesterol total, triacilgliceróis, polifenóis totais do soro, fígado e fezes. No soro e fígado foram dosados vitamina E e hidroperóxidos lipídicos. No fígado e fezes foram analisados o perfil de ácidos graxos. Coração e aorta foram processados histologicamente e feita a análise morfológica e morfométrica. Os tratamentos apresentaram considerável efeito hipocolesterolêmico com redução dos níveis séricos e hepáticos de colesterol. Houve aumento da excreção fecal de colesterol com a suplementação com antioxidantes. Não houve alteração nos triacilgliceróis, polifenóis e malondialdeído no tecido hepático dos animais. A formação de hidroperóxidos lipídicos mostrou uma tendência a ser menor quando os animais receberam os dois antioxidantes. Os grupos suplementados apresentaram maiores níveis de vitamina E séricos e hepáticos. No perfil de ácidos graxos não observou-se diferença estatística, contudo nota-se uma maior tendência a preservação dos ácidos graxos poliinsaturados quando houve suplementação com polifenois da uva. A análise morfológica revelou menor grau de evolução da placa dos animais que receberam [alfa]-tocoferol. Não houve diferença no tamanho da maior lesão encontrada.
