Morfologia, ultra-estrutura e imunohistoquímica do intestino médio de Triatoma vitticeps (Hemiptera, Triatominae)

Resumo: | The insects success is due, among other factors, to adaptation for different food resources. Usually, the gut of the insect is divided into foregut, midgut and hindgut. The midgut is responsible for key roles in the digestive process, the enzymes synthesis, digestion and absorption. This study describes the morphology, ultrastructure and the occurrence of regenerative and endocrine cells in different midgut regions of the hematophagous Hemiptera, Triatoma vitticeps, at different periods after blood meal. Twenty- seven adult insects of both sexes were dissected at different periods after feeding and the midguts were analyzed in light and transmission electron microscopes. One hundred fields for each midgut region (anterior, middle and posterior) for each animal and at different times after meal were used for morphometric analysis and for counting the number of nuclei of digestive, endocrine and regenerative cells. The digestive cells are characterized by the presence of apical microvilli, basal plasma membrane invaginations and central nucleus. The perimicrovillar membrane was present in all specimens analyzed. The anterior portion of the midgut showed digestive cells with lipid droplets, glycogen granules, developed basal labyrinth associated with mitochondria suggesting its role in processes of storage, fluid flux and ions exchange. The cells of middle and posterior regions of the midgut are rich in rough endoplasmic reticulum, lysosomes, vesicles and granules with different electron-densities. Moreover, cells of the posterior portion of the midgut are rich in electron-dense granules that are similar to hemozoyn and mitochondria located at the base of microvilli suggesting its participation in blood digestion and absorption of nutrients. The similarity between the midgut of T. vitticeps with other Hemiptera suggests that they arose from a common ancestor and that the morphological features of the midgut is more related to monophyletic origin than the type of food. The results showed in the midgut occurrence of endocrine cells characterized as the type open and positive for FMRFamide. They also show a possible association between the food cycle, sex and region of the midgut with the frequency of endocrine, digestive and regenerative cells in this Triatominae. |

Este sucesso se deve, entre outros fatores, a adaptação a diferentes hábitos alimentares. Geralmente, o intestino dos insetos é dividido em intestino anterior, médio e posterior. O intestino médio é responsável por funções fundamentais no processo digestivo, como a síntese e secreção de enzimas, digestão e absorção de alimentos. Este trabalho descreve a morfologia, a ultra- estrutura e a ocorrência de células regenerativas e células endócrinas nas diferentes regiões do intestino médio do hemíptero hematófago Triatoma vitticeps em diferentes períodos após repasto sanguíneo. Vinte e sete insetos em diferentes estádios de desenvolvimento e de ambos os sexos tiveram os intestinos médios dissecados em diferentes períodos após a alimentação e foram avaliados em microscópios de luz e eletrônico de transmissão, bem como submetidos ao teste imunohistoquímico com anti-FMRFamida para evidenciação de células endócrinas. Cem campos para cada região do intestino médio de cada animal e em períodos diferentes após repasto foram utilizados para as análises morfométricas e para a contabilização do número de núcleos das células digestivas, das células endócrinas e das células regenerativas. As células digestivas se caracterizavam pela presença de microvilosidades apicais e invaginações basais além de um núcleo central desenvolvido. A membrana perimicrovilar já estava presente em todos os espécimens analisados. A porção anterior do intestino médio apresenta as células digestivas com gotas lipídicas, grânulos de glicogênio, labirinto basal desenvolvido associado a mitocôndrias sugerindo seu papel em processos de armazenamento e transporte de fluidos e íons. As células das regiões mediana e posterior do intestino médio são ricas em retículo endoplasmático rugoso, lisossomos, vesículas e grânulos com diferentes elétron-densidades. Além disso, as células da porção posterior do intestino médio são ricas em grânulos elétron-densos que se assemelham a hemozoína e mitocôndrias dispostas nas bases de microvilosidades sugerindo sua participação na digestão de sangue e absorção de nutrientes. A semelhança entre o intestino médio do T. vitticeps com outros hemípteros sugere que estes surgiram a partir de um ancestral comum e que as características morfológicas do intestino médio relacionam-se mais a origem monofilética do que com o tipo de alimentação. Os resultados evidenciaram a presença de células enteroendócrinas do tipo aberta e positivas para FMRFamida no intestino médio de T. vitticeps.
