Resumo: | The expansion of soybean cultivation in Brazil has been accompanied by increases in occurrence of diseases and pests of this species, with these factors now limiting the obtainment of higher crop yields. Amongst the pathogens that attack soybean root knot nematodes are notable for occurring throughout the country and causing serious crop damage. Studies aiming to improve our understanding of plant-nematode interactions have been carried out but there remains a lack of information in this area. The analysis of proteins by 2D-electrophoresis associated with mass spectrometry is a well established and refined technique that can be used to investigate the molecular aspects of plant-pathogen interactions. The objective of this work was therefore to identify proteins in soybean roots that are differentially expressed in response to inoculation with root knot nematodes (Meloidogyne javanica and Meloidogyne incognita). Samples of roots from the resistant Conquista variety were evaluated which had been collected 12, 24 and 48 hours after inoculation and roots from non-inoculated plants were used as a control. Proteins were extracted using the phenol-SDS method and separated by 2D electrophoresis. Differentially expressed proteins were then detected by comparative analysis of the gels and identified using mass spectrometry (MS/MS). Various proteins/enzymes associated with cell signalling (annexin), metabolism of reactive oxygen species (ascorbate peroxidase, glutathione transferase), production of the plant hormones ethylene and jasmonic acid (protein related to maturation, the precursor of the 31kDa glycoprotein stem), lignin biosynthesis (Caffeoyl-CoA methyltransferase), amino acid metabolism (glutamine synthase, glutamate dehydrogenase, methionine synthase), redox processes (flavoprotein WrbA, isoflavone reductase) and energy metabolism (triose phosphate isomerase, enolase, UGPase, aconitate hydratase, transketolase, 6-phosphogluconate dehydrogenase, phosphoglycerate kinase, phosphoglucomutase, ATP synthase alpha subunit). These results will help to clarify the mechanisms of soybean defence against root knot nematodes and identify candidate genes that may be used in the development of transgenic plants with resistance to nematodes. |

Dentre os patógenos que acometem a cultura da soja, destacam-se os nematoides formadores de galhas por estarem presentes em todo o país e causarem sérios prejuízos à cultura. Estudos visando o entendimento da resposta da interação planta x nematoide têm sido realizados, porém, ainda há poucas informações a esse respeito. A análise de proteínas por eletroforese bidimensional associada à espectrometria de massa é uma técnica que tem sido bastante aprimorada e pode ajudar a elucidar aspectos moleculares da interação planta-patógeno. Neste sentido, objetivou-se com este trabalho identificar proteínas diferencialmente expressas em raízes de soja em resposta à inoculação com nematoides de galha (Meloidogyne javanica e Meloidogyne incognita). Foram avaliadas amostras de raízes de soja, da cultivar resistente Conquista, coletadas 12, 24 e 48 horas após a inoculação tendo como controle raízes de plantas não inoculadas. As proteínas foram extraídas pelo método fenol-SDS e separadas por eletroforese bidimensional. Por meio da análise comparativa dos géis foram detectadas proteínas diferencialmente expressas, que foram posteriormente identificadas por espectrometria de massa (MS/MS). Foram identificadas várias proteínas/enzimas que estão associadas a processos de sinalização celular (anexina), metabolismo de espécies reativas de oxigênio (ascorbato peroxidase 2, glutationa transferase), proteínas envolvidas com a produção de hormônios vegetais como etileno e ácido jasmônico (proteína relacionada à maturação, precursor da glicoproteína de 31kDa do caule), biossíntese de lignina (cafeoil-CoA metiltransferase), metabolismo de aminoácidos (glutamina sintetase, glutamato desidrogenase, metionina sintase) processo de oxido-redução (flavoproteíanWrbA, isoflavona redutase) e metabolismo energético (triose fosfato isomerase, enolase, UGPase, aconitato hidratase, transcetolase, 6 fosfogluconato desidrogenase, fosfoglicerato quinase, fosfoglicomutase, ATP sintase subunidade alfa).
