Resumo: Every year appears, in the world, thousands of new cases of cancer in human beings. In Brazil, he/she is considered more than 470.000 new cases for the year of 2006. Studies tell that many natural products are studied as agents quimioprotetors against several cancers of world occurrence. The flavonoids exercise beneficial effects in the prevention of the cancer, due to ability to inhibit the cellular cycle, the cellular proliferation, oxidative stress and to induce detoxification of enzymes and apoptose. The discovery of the transplantation of the tumors favored the study of the behavior of several cancers. The Ehrlich.ascitic tumor (TAE) it was the model used in that study in mice Swiss, inoculated with the tumor cells by road intraperitoneal, to evaluate the effect of the flavonóides quercetin, naringenin and ipriflavon in the prevention of the emergence of that tumor, as well as the quercetin and ipriflavon to treat the mice with TAE and to impede the growth of the cells of that tumor in vitro. The mice were treated with 10mg/kg of the flavonoids by 20 days. In the 21o day, the animals were inoculated by road intraperitoneal with 1,66x104 cells in PBS pH 7,4. In the experiment that sought the treatment of the tumor, the inoculation of the cells it preceded the daily use of the flavonóides quercetin and ipriflavon. After 18 days of inoculation of the cells, it was collected blood of the animals and all liquid ascítico formed in the abdominal cavity. The levels of total proteins, of the enzymes aspartato aminotransferase (AST) and alanina aminotranferase (ALT) and albumin was measured using enzymatic kits and for count of the cells it used Hemocitometro of Newbauer using the exclusion technique for the tripan blue. In the experiment that aimed at the prevention, increase was observed in the medium values of total proteins of the animals using the naringenin; reduction of the activity of AST when used the ipriflavon and of ALT when associated the quercetin + naringenin. The use of the quercetin, naringenin and quercetin+naringenin provoked an increase in the medium values of albumin. It happened a reduction in the viability of the cells present tumorais in the liquid ascítico with the use of all the tested flavonóides. The animals with TAE treated with the quercetin they presented increase in the activity of AST and in the albumin concentration. When agreements with the ipriflavon, there was a reduction in the activity of ALT. With relationship to the viability of the cells tumorais, there was a reduction, independent of the used flavonóides. The studies in vitro showed that with the increase of the concentration of the flavonóides quercetin and ipriflavon there was a smaller cellular viability. And when the animals were previously inoculated with the cells of TAE treated with 100µM of quercetin and ipriflavon, there was an increase in the medium values of total proteins and albumin in the sanguine serum of the animals and a reduction in the activity of the enzymes AST and ALT when the inoculated cells were treated with the quercetin. The flavonóides quercetin and naringenin possess an activity significant antitumoral in agreement with the criterion of NCI, where it is ended that your use as preventive it should be indicated.

No Brasil, estima-se para o ano de 2006 mais de 470.000 novos casos. Estudos relatam que muitos produtos naturais são estudados como agentes quimioprotetores contra vários cânceres de ocorrência mundial. Os flavonóides exercem efeitos benéficos na prevenção do câncer, devido a habilidade de inibir o ciclo celular, a proliferação celular, estrese oxidativo e induzir detoxificação de enzimas e apoptose. A descoberta da transplantação dos tumores favoreceu o estudo do comportamento de vários cânceres. O tumor ascítico de Ehrlich (TAE) foi o modelo utilizado nesse estudo em camundongos Swiss, inoculados com as células de tumor por via intraperitoneal, para avaliar o efeito dos flavonóides quercetina, naringenina e ipriflavona na prevenção do aparecimento desse tumor, bem como a quercetina e ipriflavona para tratar os camundongos com TAE e impedir o crescimento das células desse tumor in vitro. Os camundongos foram tratados com 10mg/kg dos flavonóides por 20 dias. No 21o dia, os animais foram inoculados por via intraperitoneal com 1,66x104 células em PBS pH 7,4. No experimento que visava o tratamento do tumor, a inoculação das células precedeu o uso diário dos flavonóides quercetina e ipriflavona. Após 18 dias de inoculação das células, foi coletado sangue dos animais e todo líquido ascítico formado na cavidade abdominal. Os níveis de proteínas totais, das enzimas aspartato aminotransferase (AST) e alanina aminotranferase (ALT) e albumina foram medidos utilizando kits enzimáticos e para contagem das células utilizou Hemocitômetro de Newbauer usando a técnica de exclusão pelo azul de tripano. No experimento que objetivava a prevenção, observou-se aumento nos valores médios de proteínas totais dos animais usando a naringenina; redução da atividade de AST quando usada a ipriflavona e de ALT quando associada a quercetina + naringenina. O uso da quercetina, naringenina e quercetina+naringenina provocaram um aumento nos valores médios de albumina. Ocorreu uma redução na viabilidade das células tumorais presentes no líquido ascítico com o uso de todos os flavonóides testados. Os animais com TAE tratados com a quercetina apresentaram aumento na atividade de AST e na concentração de albumina. Quando tratados com a ipriflavona, houve uma redução na atividade da ALT. Quanto à viabilidade das células tumorais, houve uma redução, independente dos flavonóides usados. Os estudos in vitro mostraram que com o aumento da concentração dos flavonóides quercetina e ipriflavona houve uma menor viabilidade celular. E quando os animais foram inoculados com as células do TAE previamente tratadas com 100µM de quercetina e ipriflavona, houve um aumento nos valores médios de proteínas totais e albumina no soro sanguíneo dos animais e uma redução na atividade das enzimas AST e ALT quando as células inoculadas foram tratadas com a quercetina.
