Efeito da infusão dos frutos de Momordica charantia L. em ratas diabéticas

Resumo: The objective of this study was to characterize the clinical alterations on laboratory female rats with alloxanic diabetes and on healthy female rats, treated with aqueous infusion of powdered fruits of Momordica charantia. In the biological assays, 28 female rats were induced to the experimental diabetes though intraperitoneal injection in a dose of 75 mg/kg.. The glycemia was verified on the sixth day after the induction and the animal with glycemia superior to 180 mg/dL were distributed in two groups: Treated Diabetic Group (8 animals) and Control Diabetic Group (7 animals). After dividing this two groups of female rats (healthy and diabetic rats), two others groups of healthy animals were formed: Control Health Group and Treated Healthy Group, which one with 7 animals. Treated animals were received a diary dose of 1 mL/250g of corporal weigh of the 10 % prepared infusion, and the groups-control animals, 1mL of physiologic serum, for 25 days. During the experiment were studied the following clinical parameters: weigh, hydric ingestion, alimentary ingestion, diuresis and general state. On the final of the experiment, the animals were euthanized and the following laboratorial dosages were realized: casual glicemy, hemoglobin, A1c ,insulin, C peptide and glucose-dependent insulinotropic polypeptide (GIP). Was verified weight increase among all animals of all groups and a bigger hydric and alimentary consumption in the groups of diabetic animals. After the treatment with aqueous infusion of powdered fruits of Momordica charantia, the glycemic level of treated diabetic animals was reduced to 12,4% and the treated healthy animals was reduced to 0,38%. The found values to hemoglobin A1c of diabetic animals were similar to values that are considered normal. Plasmatic concentrations to the insulin were smaller in diabetic female rats than in healthy female rats. Sanguine tenors of the C peptide of diabetic animals showed values a little smaller than healthy animals and plasmatic concentration of the glucose-dependent insulinotropic polypeptide (GIP) were shown 101,3% larger in treated diabetic animals than in no-treated diabetic animals and 68,0% above of the found values for animals of healthy control group. These results reveal that the aqueous infusion of powdered fruits of Momordica charantia is capable to promote and increase plasmatic levels of the glucose- dependent insulinotropic polypeptide, promoting a decrease of glycemic levels of animals with alloxanic diabetes.

Nos ensaios biológicos, 28 ratas foram induzidas ao diabetes experimental por via intraperitoneal na dose de 75 mg/kg. A glicemia foi verificada no sexto dia após a indução e os animais com glicemia superior a 180 mg/dL foram distribuídos em dois grupos: Grupo Diabético Tratado (8 animais) e Grupo Diabético Controle (7 animais). A partir de animais sadios, outros dois grupos foram formados: Grupo Sadio Controle e Grupo Sadio Tratado, cada um com 7 animais. Os tratados receberam dose diária de 1 mL/250g de peso corporal da infusão preparada a 10%, e os animais dos grupos-controle, 1 mL de soro fisiológico, por um período de 25 dias. Durante o experimento foram estudados os seguintes parâmetros clínicos: peso, ingestão hídrica, ingestão alimentar, diurese e estado geral. Ao final do experimento, os animais foram eutanasiados e as seguintes dosagens laboratoriais foram realizadas: glicemia casual, hemoglobina glicosilada, insulina, peptídeo C e peptídeo insulinotrópico glicose-dependente (GIP). Verificou-se um aumento do peso entre os animais de todos os grupos e um maior consumo hídrico e alimentar nos grupos de animais diabéticos. Após o tratamento com a infusão aquosa do pó dos frutos de Momordica charantia, o nível glicêmico dos animais diabéticos tratados reduziu em 12,4% e o dos animais sadios tratados reduziu em 0,38%. Os valores encontrados para a hemoglobina glicada dos animais diabéticos são próximos aos considerados normais. As concentrações plasmáticas para a insulina foram menores nas ratas diabéticas do que nas ratas sadias. Os teores sanguíneos do peptídeo C dos animais diabéticos mostraram valores pouco menores do que os animais sadios e as concentrações plasmáticas do peptídeo insulinotrópico glicose-dependente (GIP) mostraram-se 101,3% maiores nos animais diabéticos tratados do que nos animais diabéticos não-tratados e 68,0% acima dos valores encontrados para os animais do grupo controle sadio.
